Novel PLCZ1 mutation caused polyspermy during in vitro fertilization
Asian J Androl. 2024 Mar 5. doi: 10.4103/aja202376. Online ahead of print.ABSTRACTFailure of oocyte activation, including polyspermy and defects in pronuclear (PN) formation, triggers early embryonic developmental arrest. Many studies have shown that phospholipase C zeta 1 (PLCZ1) mutations cause failure of PN formation following intracytoplasmic sperm injection (ICSI); however, whether PLCZ1 mutation is associated with polyspermy during in vitro fertilization (IVF) remains unknown. Whole-exome sequencing (WES) was performed to identify candidate mutations in couples with primary infertility. Sanger sequencing was used to ...

Thyroid-stimulating hormone levels are associated with estradiol levels and impact reproductive outcomes in preconceptionally euthyroid women undergoing their first IVF/ICSI cycles
CONCLUSION(S): Serum TSH levels change significantly during COH and are associated with significant changes in estradiol levels. However, euthyroid women with high-normal TSH levels showed similar development potential for inseminated embryos and early reproductive outcomes compared to those with low TSH levels.PMID:38443586 | DOI:10.1007/s42000-024-00538-x (Source: Hormones)

Assessment of artificial intelligence model and manual morphokinetic annotation system as embryo grading methods for successful live birth prediction: a retrospective monocentric study
CONCLUSION: The developed artificial intelligence tools should be implemented in clinical practice in conjunction with the conventional morphological assessment for the conduction of optimized embryo transfer in terms of a successful live birth.PMID:38443941 | DOI:10.1186/s12958-024-01198-7 (Source: Reproductive Biology)

The effect of β-cell dysfunction on reproductive outcomes of PCOS undergoing IVF or ICSI embryo transfer cycles: a retrospective cohort study
Conclusionsβ-cell dysfunction is independently associated with increased miscarriage rate and decreased live birth rate in women with PCOS. It also plays a synergistic role with IR in terms of the reproductive outcomes, while the influence of IR overweighs that of β-cell dysfunction. (Source: Frontiers in Endocrinology)

Establishment and validation of a nomogram for subsequent first-cycle live births in patients diagnosed with recurrent implantation failure: a population-based analysis
ConclusionWe have developed and validated a novel predictive model that estimates a woman’s chances of having a live birth after a diagnosis of RIF and provides clinicians with a personalized clinical decision-making tool. (Source: Frontiers in Endocrinology)

Influx of zwitterionic buffer after intracytoplasmic sperm injection (ICSI) membrane piercing alters the transcriptome of human oocytes
ConclusionThe influence of zwitterionic buffer on intrinsic cellular mechanisms provides numerous concerns for their use in IVF clinical applications. The primary concern is the ICSI procedure, in which the surrounding buffer is allowed influx into the oocytes after membrane piercing.  Selecting a physiological bicarbonate buffer may reduce imposed stress on oocytes, resulting in improved embryo development and clinical results because intracellular MOPS, and especially HEPES, may negatively impact intrinsic biological mechanisms, as revealed by transcriptome changes. These find ings further support the utilization of bic...

Achieving an optimal pregnancy outcome through the combined utilization of micro-TESE and ICSI in cryptorchidism associated with a non-canonical splicing variant in RXFP2
ConclusionThis is the first cryptorchidism case carrying a novel causative non-canonical splicingRXFP2 variant. The combined approach of micro-TESE and ICSI contributed to an optimal pregnancy outcome. Our literature review demonstrated thatRXFP2 variants caused cryptorchidism in a recessive inheritance pattern, rather than a dominant pattern. (Source: Journal of Assisted Reproduction and Genetics)

GSE241089 First cleavage is a manifestation of the geometry of the unfertilized oocyte: implications for monozygotic twinning in mice.

A hitherto untested hypothesis is that allocation depends on the orientation of 1st cleavage. However, the orientation is influenced by the site of sperm entry in the oocyte, which can be almost anywhere on the membr...
