Genes, Vol. 15, Pages 53: Exploring the Regulation and Function of Rpl3l in the Development of Early-Onset Dilated Cardiomyopathy and Congestive Heart Failure Using Systems Genetics Approach
Conclusions: RPL3L is highly expressed in the heart tissue of humans and mice. Expression of Rpl3l and its upstream regulators, Myl4 and Sdha, correlate with multiple cardiac function traits in murine GRPs of BXD mice, while RPL3L and SDHA correlate with immune cell infiltration in DCM patient hearts, suggesting important roles for RPL3L in DCM and CHF pathogenesis via immune inflammation, necessitating experimental validations of Myl4 and Sdha in Rpl3l regulation. Mechanical Dispersion Is Associated With Ventricular Arrhythmias and Sudden Cardiac Death in Nonischemic Dilated Cardiomyopathy
Patients with nonischemic dilated cardiomyopathy (NIDCM) are at higher risk of ventricular arrhythmias (VAs) and sudden cardiac death (SCD), but the use of implantable cardioverter-defibrillators (ICDs) is not recommended in all-comers.1 Risk stratification is paramount when deciding ICD implantation in primary prevention before VAs occur. Although late gadolinium enhancement (LGE) using cardiac magnetic resonance (CMR) has been demonstrated to be strongly associated with VA during follow-up,2 CMR is not widely available for repeated assessments and myocardial substrate may vary with time. Striated preferentially expressed gene deficiency leads to mitochondrial dysfunction in developing cardiomyocytes
AbstractA deficiency of striated  preferentially expressed gene (Speg), a member of the myosin light chain kinase family, results in abnormal myofibril structure and function of immature cardiomyocytes (CMs), corresponding with a dilated cardiomyopathy, heart failure and perinatal death. Mitochondrial development plays a role in cardiomyocyte maturation. Therefore, this study investigated whetherSpeg deficiency ( – / – ) in CMs would result in mitochondrial abnormalities.Speg wild-type andSpeg−/− C57BL/6 littermate mice were utilized for assessment of mitochondrial structure by transmission electron and confocal m...

Generation of an RBM20-mutation-associated left-ventricular non-compaction cardiomyopathy iPSC line (UMGi255-A) into a DCM genetic background to investigate monogenetic cardiomyopathies
Stem Cell Res. 2023 Dec 16;74:103290. doi: 10.1016/j.scr.2023.103290. Online ahead of print.ABSTRACTRBM20 mutations account for 3 % of genetic cardiomypathies and manifest with high penetrance and arrhythmogenic effects. Numerous mutations in the conserved RS domain have been described as causing dilated cardiomyopathy (DCM), whereas a particular mutation (p.R634L) drives development of a different cardiac phenotype: left-ventricular non-compaction cardiomyopathy. We generated a mutation-induced pluripotent stem cell (iPSC) line in which the RBM20-LVNC mutation p.R634L was introduced into a DCM patient line with rescued RB...

Extracellular vesicles (EVs): A promising therapeutic tool in the heart tissue regeneration
Extracellular vesicles paracrine effects play a primary role in the regenerative ability. EVs and the bioactive molecules contained in the human dental pulp stem cells influence the differentiation process toward cardiomyogenic lineage. Differentiation of dental pulp stem cells into cardiomyocytes induced by EVs derived modulating the differentiation process toward cardiomyogenic lineage. AbstractMesenchymal stem cells (MSCs) treatment has been widely explored as a therapy for myocardial infarction, peripheral ischemic vascular diseases, dilated cardiomyopathy, and pulmonary hypertension. Latest in vitro studies suggest th...
