Efficacy and safety of eszopiclone combined with drug therapy in the treatment of insomnia after stroke: A network meta-analysis and systematic review
ConclusionMulti-drug therapy based on eszopiclone can effectively improve the sleep quality of patients with insomnia after stroke, and ESZ+SDOL has significant efficacy and safety. However, due to the limitations of this study, efficacy ranking cannot fully explain the superiority or inferiority of clinical efficacy. In the future, more multicentre, large sample, double-blind randomized controlled trials are needed to supplement and demonstrate the results of this study. (Source: PLoS One)

Prazosin Enhances the Effectiveness of Mirtazapine on Anxiety- and Depression-Like Behaviors in Rats during Cocaine Withdrawal
This study sought to determine whether chronic dosing of mirtazapine plus prazosin during cocaine withdrawal reduced depression- and anxiety-like behaviors that characterize cocaine withdrawal inWistar rats. Cocaine-pre-treated Wistar rats were subjected to a 60-day cocaine withdrawal period during which depression- and anxiety-like behaviors were evaluated in open field tests (OFT), the elevated plus-maze (EPM), the light –dark box test (LDT), the forced swimming test (FST), and spontaneous locomotor activity (SLA). We found 1) that chronic dosing of mirtazapine (30 mg/kg) + prazosin (1 mg/kg) decreased depression...

Inpatient Management of Hyperemesis Gravidarum
Obstet Gynecol. 2024 Feb 1. doi: 10.1097/AOG.0000000000005518. Online ahead of print.ABSTRACTHyperemesis gravidarum has a reported incidence of approximately 0.3-3% of pregnancies. Without treatment, refractory hyperemesis gravidarum can result in dehydration, electrolyte deficiencies, and severe nutritional deficiencies, resulting in significant maternal morbidity. The overall goals of inpatient management of refractory hyperemesis gravidarum are the resumption of oral intake to an adequate level to maintain hydration and nutrition, including the ability to tolerate oral pharmacotherapy. Patients initially are stabilized ...

Inhibitory Actions of Antidepressants, Hypnotics, and Anxiolytics on Recombinant Human Acetylcholinesterase Activity
Biol Pharm Bull. 2024;47(1):328-333. doi: 10.1248/bpb.b23-00719.ABSTRACTAlzheimer's disease (AD) is accompanied by behavioral and psychological symptoms of dementia (BPSD), which is often alleviated by treatment with psychotropic drugs, such as antidepressants, hypnotics, and anxiolytics. If these drugs also inhibit acetylcholinesterase (AChE) activity, they may contribute to the suppression of AD progression by increasing brain acetylcholine concentrations. We tested the potential inhibitory effects of 31 antidepressants, 21 hypnotics, and 12 anxiolytics on recombinant human AChE (rhAChE) activity. At a concentration of 1...

Molecules, Vol. 29, Pages 676: Direct Immersion & ndash;Solid Phase Microextraction for Therapeutic Drug Monitoring of Patients with Mood Disorders
This article discusses a new method for monitoring drug concentrations in blood samples from patients with mood disorders. The method uses solid-phase microextraction to extract analytes directly from blood samples. It has been adapted to identify the most commonly used drugs in mood disorders, including amitriptyline, citalopram, fluoxetine, paroxetine, sertraline, trazodone, duloxetine, venlafaxine, lamotrigine, quetiapine, olanzapine, and mirtazapine. The analysis is carried out using high-performance liquid chromatography coupled with mass spectroscopy. The proposed DI-SPME/LC-MS method allows for a simple and quick sc...

Medicating the coast in a metropolitan city: Enantiomeric profiles and joint probabilistic risk assessment of antidepressants and antihistamines
Environ Int. 2024 Jan 9;184:108434. doi: 10.1016/j.envint.2024.108434. Online ahead of print.ABSTRACTPharmaceuticals are receiving increasing attention as emerging contaminants in the aquatic environment. Herein, we investigated the occurrence of 11 antidepressants, 6 antihistamines and 4 metabolites in treated wastewater effluents, rivers, stormwater, and seawater in Hong Kong, with special focus on chirality. The average levels of ∑pharmaceuticals ranged from 0.525 to 1070 ng/L in all samples and the total annual mass load of target pharmaceuticals in the marine environment of Hong Kong was 756 kg/y. Antihistamines acc...

Pharmacokinetics of a Single Transdermal Dose of Mirtazapine in Rhesus Macaques ( < em > Macaca mulatta < /em > )
Comp Med. 2023 Dec 1;73(6):432-438. doi: 10.30802/AALAS-CM-23-000060.ABSTRACTDecreased appetite is a common clinical problem in captive rhesus macaques (Macaca mulatta). Mirtazapine, a tetracyclic antidepressant originally developed for humans, has shown promise as a safe and effective promoter of weight gain and appetite in several veterinary species including rhesus and cynomolgus macaques. Although mirtazapine is available as oral formulations, transdermal delivery in macaques with reduced appetite would allow quick, painless, topical application. Here we describe the pharmacokinetics of a single application of a widely...

Age of onset for increased dose-adjusted serum concentrations of antidepressants and association with sex and genotype: An observational study of 34,777 individuals
ConclusionThe age-related increase in C/D ratio starts in middle-aged adults and progresses up to more than twofold higher C/D ratio in the oldest old. Sertraline seems to be less prone to age-related changes in C/D ratio than the other antidepressants. (Source: European Journal of Clinical Pharmacology)
