A potent neutralizing nanobody targeting a unique epitope on the receptor-binding domain of SARS-CoV-2 spike protein
In this study, we reported the identification and structural biological characterization of a nanobody from a RBD-immunized alpaca. The nanobody, termed as 2S-1-19, shows outstanding neutralizing activity against both pseudotyped and authentic SARS-CoV-2 viruses. The crystal structure of 2S-1-19 bound to SARS-CoV-2 RBD reveals an epitope that overlaps with the binding site for ACE2. We also showed that 2S-1-19 reserves promising, though compromised, neutralizing activity against the Delta variant and that the trivalent form of 2S-1-19 remarkably increases its neutralizing capacity. Despite this, neither the monomeric or tr...

Epidemiological and genetic characteristics of foot-and-mouth disease virus in China from 2010 to 2022
This study summarized 123 cases of FMD reported in China between 2010 and 2022, using data from the official website of the Chinese Center for Animal Disease Control and Prevention. The epidemic situation and genetic characteristics of FMDV in China were studied through phylogenetic analysis, amino acid variation analysis of antigenic epitopes, and genetic recombination analysis. The findings provide important references for predicting the FMDV epidemic situation in China, developing vaccines, and effectively preventing and controlling FMD.PMID:37984153 | DOI:10.1016/j.virol.2023.109940 (Source: Virology)

A cell-based assay to discover inhibitors of Zika virus RNA-dependent RNA polymerase
Virology. 2023 Nov 11;589:109939. doi: 10.1016/j.virol.2023.109939. Online ahead of print.ABSTRACTZika virus (ZIKV) belongs to Flaviviridae, the Flavivirus genus. Its infection causes congenital brain abnormalities and Guillain-Barré syndrome. However, there are no effective vaccines, no FDA-approved drugs to manage ZIKV infection. The non-structural protein NS5 of ZIKV has been recognized as a valuable target of antivirals because of its RNA-dependent RNA polymerase (RdRp) and methyltransferase (MTase) activities essential for viral RNA synthesis. Here, we report a cell-based assay for discovering inhibitors of ZIKV NS5 ...

Inhibitory effects of quercetin on porcine epidemic diarrhea virus in vitro and in vivo
This study aimed to explore the therapeutic effects of quercetin in PEDV infections in vitro (Vero cells) and in vivo (suckling piglets). Using transmission electron microscopy and laser confocal microscopy, we found that PEDV infection promotes the accumulation of lipid droplets (LDs). In vitro, studies showed that quercetin inhibits LD accumulation by down-regulating NF-κB signaling and IL-1β, IL-8, and IL-6 levels, thereby inhibiting viral replication. In vivo, studies in pigs demonstrated that quercetin can effectively relieve the clinical symptoms and intestinal injury caused by PEDV. Collectively, our findings sugg...
