Luteolin ameliorates loperamide-induced functional constipation in mice
This study aimed to explore the role of luteolin and its functional mechanism in loperamide-induced FC mice. Our results showed that luteolin treatment reversed the reduction in defecation frequency, fecal water content, and intestinal transit ratio, and the elevation in transit time of FC models. Consistently, luteolin increased the thickness of the muscular layer and lessened colonic histopathological injury induced by loperamide. Furthermore, we revealed that luteolin treatment increased the expression of neuronal protein HuC/D and the levels of intestinal motility-related biomarkers, including substance P (SP), vasoact...

Determination of Region-Specific Roles of the M3 Muscarinic Acetylcholine Receptor in Gastrointestinal Motility
ConclusionsThis study provided the first direct evidence that the M3 receptor has different region-specific roles in the motility function between the small intestine and colon in physiological and pathophysiological contexts. Selective PAMs designed for targeted subtypes of muscarinic receptors are useful for elucidating the subtype-specific function. (Source: Digestive Diseases and Sciences)

COVID-19 –associated colitis
A 63-year-old man was admitted to the hospital because of COVID-19 pneumonia and was intubated a few days later because of respiratory failure. Soon afterward, he experienced profuse watery diarrhea. The result of stool analysis was negative for viral and bacterial pathogens, and Clostridium difficile and cytomegalovirus infections were ruled out. Therapy with loperamide was initiated; however, the symptoms persisted. The patient underwent colonoscopy 21 days after the start of the symptoms. As part of the COVID-19 treatment, he was receiving methylprednisolone with dosage tapering. (Source: Gastrointestinal Endoscopy)

Final findings from the CONTROL trial: Strategies to reduce the incidence and severity of neratinib-associated diarrhea in patients with HER2-positive early-stage breast cancer
CONCLUSIONS: These complete results from CONTROL show improved neratinib tolerability with proactive management at the start of therapy. Two-week neratinib DE with loperamide as needed was particularly effective.GOV REGISTRATION NUMBER: NCT02400476.PMID:36702070 | DOI:10.1016/j.breast.2022.12.003 (Source: Breast)
