Racecadotril versus Loperamide in acute radiation enteritis: a randomized, double-blind, phase 3, non-inferiority trial.
There is currently no gold standard available for the management of acute radiation enteritis. We compared the efficacy and safety of Racecadotril an antihypersecretory drug versus Loperamide an anti-motility agent in acute radiation enteritis. Zhishi Daozhi decoction alleviates constipation induced by a high-fat and high-protein diet via regulating intestinal mucosal microbiota and oxidative stress
ConclusionConstipation induced by HFHPD can increase pathogenic bacteria in the intestinal mucosa, while ZDD can effectively relieve constipation, reduce the relative abundance of pathogenic bacteria, and alleviate oxidative stress injury. In addition, high-dose ZDD can increase the abundance of beneficial bacteria, which is more conducive to the treatment of constipation. (Source: Frontiers in Microbiology)

Low anterior resection syndrome
Ann Gastroenterol Surg. 2023 May 23;7(5):719-724. doi: 10.1002/ags3.12695. eCollection 2023 Sep.ABSTRACTLow anterior resection syndrome (LARS) is the distressful defecatory functional problem after sphincter-saving surgery for rectal cancer. Although the symptoms of fecal urgency, frequency, and incontinence may develop in most of the patients after surgery, there is no definitive treatments for LARS. Multifactorial etiologies and various risk factors have been identified, but the reduction of storage capacity in the rectum is one of the main reasons for LARS. Anal sphincter muscle or nerve damage during rectoanal resectio...

Modulation of gut microbiota ecosystem by a glucan-rich snail mucin heteropolysaccharide attenuates loperamide-induced constipation
Int J Biol Macromol. 2023 Aug 26:126560. doi: 10.1016/j.ijbiomac.2023.126560. Online ahead of print.ABSTRACTThe present study aimed to investigate the effect of oral administration of snail-derived mucin extract (SM) on ameliorating constipation symptoms of loperamide-induced constipated rats (n = 6). The analytical results indicated that SM mainly contains a glucan-rich snail mucin heteropolysaccharide with high molecular weights (108.5-267.9 kDa), comprising primarily of glucose (64.9 %) and galactose (22.4 %) with some deoxyhexoses (5.0 %) and hexosamines (4.9 %). Daily SM administration at doses of 10-40 mg/kg/day to t...

Thin-film nanocomposite membranes for efficient removal of emerging pharmaceutical organic contaminants from water
Environ Res. 2023 Aug 17:116905. doi: 10.1016/j.envres.2023.116905. Online ahead of print.ABSTRACTMembranes are receiving significant attention to remove emerging organic micropollutants (OMPs) from wastewater and natural water sources. Herein, we report the facile preparation of a novel thin-film nanocomposite (TFN) membrane with high permeability and efficient removal of OMPs. ZnO nanoparticles were first synthesized using the co-precipitation method and functionalized with N1-(3-Trimethoxysilylpropyl)diethylenetriamine to make the surface rich with amine groups and then synthesized nanomaterials were covalently cross-li...

Neratinib + fulvestrant + trastuzumab for HR-positive, HER2-negative, HER2-mutant metastatic breast cancer: outcomes and biomarker analysis from the SUMMIT trial
CONCLUSIONS: The benefit of N+F+T for HR+ HER2-mutant MBC after progression on CDK4/6 inhibitors is clinically meaningful and, based on this study, N+F+T has been included in National Comprehensive Cancer Network treatment guidelines. SUMMIT has improved our understanding of the translational implications of targeting HER2 mutations with neratinib-based therapy.PMID:37597578 | DOI:10.1016/j.annonc.2023.08.003 (Source: Ann Oncol)

Discovery of novel carboxylesterase 2 inhibitors for the treatment of delayed diarrhea and ulcerative colitis
This study identified a lead compound 1 with a novel scaffold by high-throughput screening in our in-house library. After a comprehensive structure-activity relationship study, the optimal compound 24 was discovered as an efficient and highly selective hCES2 inhibitor (hCES2: IC50 = 6.72 μM; hCES1: IC50 > 100 μM). Further enzyme kinetics study indicated that compound 24 is a reversible inhibitor of hCES2 with competitive inhibition mode (Ki = 6.28 μM). The cell experiments showed that compound 24 could reduce the level of hCES2 in living cells (IC50 = 6.54 μM). The modeling study suggested that compound 24 fitted ve...
