Glanders and brucellosis in equids from the Amazon region, Brazil
Acta Trop. 2022 Mar 27;231:106429. doi: 10.1016/j.actatropica.2022.106429. Online ahead of print.ABSTRACTGlanders and brucellosis are zoonotic infectious diseases that affect equids in several countries worldwide. On Marajó Island (Amazon region of Brazil), Marajoara and Puruca horses, which are well adapted to the climatic and territorial adversities of the region, play a fundamental role in the local economy and in the sociocultural lives of the population. However, these animals have undergone a drastic reduction in number, markedly due to precarious veterinary care, unknown causes of morbidity and mortality, and disor...

Molecular characterization of Burkholderia mallei strains isolated from horses in Brazil (2014-2017)
The objective of this work was to provide additional knowledge on the diversity of the strains circulating in Brazil. Six Burkholderia mallei isolates obtained during necropsies of glanderous horses between 2014 and 2017 in two different states (Pernambuco and Alagoas) were analyzed by polymerase chain reaction-high-resolution melting (PCR-HRM). While four strains (9902 RSC, BM_campo 1, BM_campo 3 and UFAL2) clustered in the L3B2 branch, which already includes the Brazilian 16-2438_BM#8 strain, two strains (BM_campo 2.1 and BM_campo 2.2) clustered within the L3B3sB3 branch, which mostly includes older isolates, from Europe...

Polymerase Spiral Reaction (PSR) as a novel rapid colorimetric isothermal point of care assay for detection of Trypanosoma evansi genomic DNA
Vet Parasitol. 2021 Dec 28;302:109644. doi: 10.1016/j.vetpar.2021.109644. Online ahead of print.ABSTRACTPolymerase spiral reaction (PSR) opens new avenues for specific diagnosis of pathogens known for cryptic infection at field level and its application is still unexplored in the field of parasitology. The present study aimed to explore and optimize colorimetric based PSR technique for the detection of Trypanosoma evansi in the blood of the host by targeting the 196bp Invariable Surface Glycoprotein (ISG) gene of Trypanosoma evansi. The specificity of the test was determined against Theileria equi, Theileria annulata, Babe...

Development of a sensitive competitive enzyme-linked immunosorbent assay for serodiagnosis of < i > Burkholderia mallei < /i > , a Tier 1 select agent
In this study, we raised a monoclonal antibody (mAb) against the lipopolysaccharide (LPS) ofB.mallei and developed a competitive enzyme-linked immunosorbent assay (cELISA) forB.mallei infection. Using the titrated optimal conditions ofB.mallei-LPS (2 ng) for microtiter plate coating, sample serum dilution at 1:20 and 3.5 ng/ μL anti-LPS mAb B5, the cutoff value of the cELISA was determined using serum samples from 136 glanders-free seronegative horses in Hong Kong. All calculated percentage inhibition (PI) values from these seronegative samples were below 39.6% inhibition (1.5 standard deviations above mean PI) and was us...

Genomic pattern analysis of < em > Burkholderia mallei < /em > field isolates by pulsed-field gel electrophoresis (PFGE) discriminatory typing
CONCLUSION: PFGE showed more discriminatory power and considerable reproducibility for molecular typing of B. mallei strains in our study. It is standardized the approaches for outbreak detection, pathogen phylogeny, molecular epidemiology, and population studies.PMID:34900154 | PMC:PMC8629827 | DOI:10.18502/ijm.v13i5.7419 (Source: Iranian Journal of Microbiology)
