GSE160870 Gene Expression of A375 Cells After Fluvastatin Treatment
Contributors : Wesley Y Yu ; John J Pink ; Alexander Miron ; E R ChanSeries Type : Expression profiling by high throughput sequencingOrganism : Homo sapiensDespite advances in melanoma treatment, more than 70% of patients with distant metastasis die within 5 years. Proactive treatment of early melanoma to prevent metastasis could save lives and reduce overall healthcare costs. Currently, there are no treatments specifically designed to prevent early melanoma from progressing to metastasis. We used the Connectivity Map (cMap) to conduct an in silico drug screen and identified HMGCR inhibitors (statins) as a drug class tha...

Repurposing of statins for Buruli Ulcer treatment: antimicrobial activity against Mycobacterium ulcerans
In conclusion, statins hold promise as potential treatment options for Buruli Ulcer. Further studies are necessary to validate their effectiveness and understand the mechanism of action of statins against M. ulcerans. (Source: Frontiers in Microbiology)

CaaX-motif-adjacent residues influence G protein gamma (G γ) prenylation under suboptimal conditions
We examined the prenylation of carboxy-terminus (Ct) mutated Gγ in cells exposed to Fluvastatin and prenyl transferase inhibitors and monitored the subcellular localization of fluorescently tagged Gγ subunits and their mutants using live-cell confocal imaging. Reversible optogenetic unmasking-masking of Ct residues was used to probe their contribution to prenylation and membrane interactions of the prenylated proteins. Our findings suggest that specific Ct residues regulate membrane interactions of the Gγ polypeptide, statin sensitivity, and extent of prenylation. Our results also show a few hydrophobic and charged resi...

Pravastatin for lowering lipids
CONCLUSIONS: Pravastatin lowers blood total cholesterol, LDL cholesterol and triglyceride in a dose-dependent linear fashion. This review did not provide a good estimate of the incidence of harms associated with pravastatin because of the lack of reporting of adverse effects in 48.4% of the randomized placebo-controlled trials.PMID:37721222 | DOI:10.1002/14651858.CD013673.pub2 (Source: Cochrane Database of Systematic Reviews)

Metal-Free Directed C –H Borylation of Indoles at the Sterically Congested C2 Position
Synlett DOI: 10.1055/a-2126-1750 During the past few decades, transition metal-catalyzed C–H borylation has been one of the most notable advances in synthetic chemistry and has been widely employed in the preparation of organoboron reagents. Due to economic and heavy-metal-residue concerns, there is significant interest in the development of metal-free processes to mimic metallic systems. Here, we disclose a highly efficient metal-free approach for the directed C–H borylation of C3-substituted indoles at the sterically congested C2 position that uses the inexpensive boron reagent BBr3. Compared with the conventional me...

Effect of Statins on Superoxide Dismutase Level: A Systematic Review
CONCLUSION: Our systematic review suggests that treatment with statins has a positive effect on SOD activity. However, evidence from further randomized controlled trials is required to confirm the potential antioxidant effect of statin therapy.PMID:37653630 | DOI:10.2174/0929867331666230831145809 (Source: Current Medicinal Chemistry)

Neutrophils for Revascularization Require Activation of CCR6 and CCL20 by TNF α
CONCLUSIONS: We demonstrate that site-specific activation of the CCL20-CCR6 axis via TNFα recruits proangiogenic VEGFA-expressing neutrophils to sites of ischemic injury for initiation of muscle tissue revascularization. The findings provide an attractive option for tissue revascularization, particularly under diabetic conditions.PMID:37641931 | DOI:10.1161/CIRCRESAHA.123.323071 (Source: Cell Research)
