Treatment of vismodegib-associated muscle cramps with cyclobenzaprine: A retrospective review
To the Editor: Basal cell carcinomas (BCCs) represent the most frequent cancer, caused by inappropriate activation of the hedgehog signaling pathway. While  most BCCs can be cured with surgery, some progress to locally advanced (laBCC) or metastatic (mBCC) disease. In 2012, the US Food and Drug Administration approved vismodegib, the first hedgehog pathway inhibitor, for the treatment of laBCC and mBCC,1 followed by the approval of sonidegib for treat ment of laBCC in 2015.2 (Source: Journal of the American Academy of Dermatology)

Bioequivalence of cyclobenzaprine hydrochloride extended-release capsule taken  intact or sprinkled over applesauce
.
CONCLUSION: CER capsules intact and sprinkled over applesauce are bioequivalent. Sprinkling CER capsule contents is not expected to affect efficacy or safety and can, therefore, be an option for patients with musculoskeletal pain and difficulty swallowing capsules.

CNS Summit 2017 Abstracts of Poster Presentations
Conclusion: This novel technology discriminates and quantifies subtle differences in behavior and neurological impairments in subjects afflicted with neurological injury/disease. KINARM assessments can be incorporated into multi-center trials (e.g., monitoring stroke motor recovery: NCT02928393). Further studies will determine if KINARM Labs can demonstrate a clinical effect with fewer subjects over a shorter trial period. Disclosures/funding: Dr. Stephen Scott is the inventor of KINARM and CSO of BKIN Technologies.   Multiplexed mass spectrometry assay identifies neurodegeneration biomarkers in CSF Presenter: Chelsky...

Pharmacological treatment of oro ‐facial pain – health technology assessment including a systematic review with network meta‐analysis
In conclusion, based on a limited number of studies, evidence provided with network meta‐analysis showed that clonazepam and capsaicin are effective in treatment of BMS and that the muscle relaxant cyclobenzaprine has a positive treatment effect for TMD‐muscle pain. 854. Phase 2 Multisite Double-Blind Placebo-Controlled Trial of TNX-102 Sl in Military-Related Posttraumatic Stress Disorder (PTSD): Mediators and Moderators of Treatment Response
Evidence-based pharmacotherapies are lacking for military-related PTSD. TNX-102 SL* is a proprietary formulation of cyclobenzaprine (CBP) for bedtime sublingual administration that bypasses first pass metabolism and antagonizes 5-HT2A, 1-adrenergic and histaminergic-1 receptors. It is hypothesized to improve PTSD via effects on core symptoms of sleep disturbance and hyperarousal. (Source: Biological Psychiatry)

Inclusion complexes of β -cyclodextrin with tricyclic drugs: an X-ray diffraction, NMR and molecular dynamics study
Abstract Tricyclic fused-ring cyclobenzaprine (1) and amitriptyline (2) form 1:1 inclusion complexes with β-cyclodextrin (β-CD) in the solid state and in water solution. Rotating frame NOE experiments (ROESY) showed the same geometry of inclusion for both 1/β-CD and 2/β-CD complexes, with the aromatic ring system entering the cavity from the large rim of the cyclodextrin and the alkylammonium chain protruding out of the cavity and facing the secondary OH rim. These features matched those found in the molecular dynamics (MD) simulations in solution and in the solid state from single-crystal X-ray diffrac...

Tramadol vs. Everything: 744 Drug Interactions and Counting
BY THOMAS BARROWS, MDA 43-year-old woman presented to our emergency department with complaints of anxiety, intermittent “cramping” in her left hand, and jerking movements of her body that had been going on for five days. She had depression following the death of her son 14 months earlier, for which she was started on Effexor 37.5 mg once daily two weeks earlier. One week before this visit, the patient had back pain and headache, for which she was evaluated at a local emergency department. She had lumbar x-rays showing mild degenerative changes in her spine and a normal head CT scan. The emergency physician...

Management of fibromyalgia: practical guides from recent evidence-based guidelines.
Authors: Häuser W, Ablin J, Perrot S, Fitzcharles MA Abstract Fibromyalgia (FM) is a prevalent and costly condition worldwide, affecting approximately 2% of the general population. Recent evidence- and consensus-based guidelines from Canada, Germany, Israel and the European League Against Rheumatism aim to support physicians in achieving a comprehensive diagnostic work-up of patients with chronic widespread pain (CWP) and to assist patients and physicians in shared decision making on treament options. Every patient with CWP requires at the first medical evaluation a complete history, medical examination a...

Cyclobenzaprine Raises ROS Levels in < i > Leishmania infantum < /i > and Reduces Parasite Burden in Infected Mice
Conclusion/SignificanceTo the best of our knowledge, this study is the first report of thein vivo antileishmanial activity of the FDA-approved drug CBP. Modulation of immune response and induction of oxidative stress in parasite seem to contribute to this efficacy. (Source: PLoS Neglected Tropical Diseases)
