Cinnamic acid and lactobionic acid based nanoformulations as a potential antiamoebic therapeutics
Exp Parasitol. 2023 Jan 25:108474. doi: 10.1016/j.exppara.2023.108474. Online ahead of print.ABSTRACTAcanthamoeba castellanii causes granulomatous amoebic encephalitis, an uncommon but severe brain infection and sight-threatening Acanthamoeba keratitis. Most of the currently used anti-amoebic treatments are not always effective, due to persistence of the cyst stage, and recurrence can occur. Here in this study we synthesize cinnamic acid and lactobionic acid-based magnetic nanoparticles (MNPs) using co-precipitation technique. These nanoformulations were characterized by Fourier transform infrared spectroscopy and Atomic f...

Antiamoebic Susceptibility in Acanthamoeba Keratitis: Comparison of Isolates From South India and Northern California
Conclusions: This study found no statistically significant differences in antiamoebic susceptibility of Indian versus US samples from Acanthamoeba keratitis clinical isolates. These findings suggest that differences in antiamoebic susceptibility are likely not responsible for differential outcomes in Acanthamoeba keratitis between the 2 locations. (Source: Cornea)

Occurrence and molecular characterization of Acanthamoeba, Naegleria fowleri and Blastocystis in water samples from various sources in Egypt
Acta Trop. 2022 Oct 27;237:106733. doi: 10.1016/j.actatropica.2022.106733. Online ahead of print.ABSTRACTPCR-testing coupled to isolate sequencing was conducted to detect prevalence and various genotypes/subtypes of 3 neglected waterborne protists (Acanthamoeba, Naegleria fowleri and Blastocystis) in water samples from various sources in Dakahlia governorate, Egypt. Out of 62 protozoan-suspected samples by microscopy, Acanthamoeba was molecularly confirmed in 24 (38.7%) samples from various sources including tap water. Twenty Acanthamoeba isolates were successfully-sequenced; 18 were designated as the genotype T3 and 2 as ...

Update on the Management of Acanthamoeba Keratitis
AbstractPurpose or ReviewIn this article, we review the latest diagnostic modalities and management ofacanthamoeba keratitisRecent FindingsThe incidence as well as severity ofacanthamoeba keratitis is increasing, making this a challenging disease to treat, especially in patients who have a delay in diagnosis or severe disease at presentation. Newer tools such as voriconazole, miltefosine, and collagen crosslinking may be needed for infection control. Complications such as persistent infection, severe thinning, or perforation may necessitate surgical intervention.SummaryWhile traditional therapies such as polyhexamethylene ...

Pitavastatin loaded nanoparticles: a suitable ophthalmic treatment for Acanthamoeba Keratitis inducing cell death and autophagy in Acanthamoeba polyphaga
Eur J Pharm Biopharm. 2022 Sep 23:S0939-6411(22)00215-6. doi: 10.1016/j.ejpb.2022.09.020. Online ahead of print.ABSTRACTStatins are effective sterol lowering agents with high amoebicidal activity. Nevertheless, due to their poor aqueous solubility, they remain underused especially in eye drop formulation. The aim of the present study is to develop Pitavastatin loaded nanoparticles suitable for ophthalmic administration and designed for the management of Acanthamoeba Keratitis. These nanocarriers are aimed to solve both the ophthalmic route-associated problems and the limited aqueous drug solubility issues of Pitavastatin. ...

As a monotherapy, alpha-mangostin is effective for the treatment of AK; however, its bioavailability is quite poor. Moreover, the efficacy of therapy is contingent on the parasite and virulent strains. To improve re...

Polyhexamethylene biguanide hydrochloride (PHMB)-properties and application of an antiseptic agent. A narrative review
Eur J Ophthalmol. 2022 Sep 9:11206721221124684. doi: 10.1177/11206721221124684. Online ahead of print.ABSTRACTThe prevention and management of ocular surface infections is still one of the great challenges for ophthalmologists. The spread of antimicrobial resistance makes it necessary to use antiseptic substances with a broad antimicrobial spectrum. Polyhexamethylene biguanide hydrochloride (Polyhexanide, PHMB) is a broad-spectrum antiseptic with excellent tolerance and a low-risk profile. Its physicochemical action on the phospholipid membrane and DNA replication or repair mechanism, prevents or impedes the development of...
