Simvastatin, but Not Atorvastatin, Is Associated with Higher Peak Rivaroxaban Serum Levels and Bleeding: an Asian Cohort Study from Singapore
ConclusionOur findings suggest that concomitant use of simvastatin with rivaroxaban may be associated with bleeding events in an Asian cohort. Further studies using physiologically based pharmacokinetic modelling are required to investigate the drug-drug interactions between these drugs.

PPP1R12C Promotes Atrial Hypocontractility in Atrial Fibrillation
CONCLUSIONS: Patients with AF exhibit increased levels of PPP1R12C protein compared with controls. PPP1R12C overexpression in mice increases PP1c targeting to MLC2a and causes MLC2a dephosphorylation, which reduces atrial contractility and increases AF inducibility. These findings suggest that PP1 regulation of sarcomere function at MLC2a is a key determinant of atrial contractility in AF.PMID:37737016 | DOI:10.1161/CIRCRESAHA.123.322516

The prevalence and prognosis of asymptomatic intracranial atherosclerosis in a community-based population: Results based on high-resolution magnetic resonance imaging
CONCLUSIONS: We found that ICAD detected using HR-MRI increases the long-term risk of a first-ever ischemic stroke in a stroke-free population, suggesting that the current primary prevention protocol of stroke awaits further optimization.PMID:37738517 | DOI:10.1111/ene.16057

Association Between Calcium Supplementation and the Risk of Cardiovascular Disease and Stroke: A  Systematic Review and Meta-Analysis
Some observational studies and randomised controlled trials (RCTs) have reported an association between calcium supplementation and increased risk of cardiovascular disease. Previous meta-analyses on the topic, based on data from RCTs and observational studies, have contradictory findings. This meta-analysis was conducted to determine the difference in associated risks of calcium supplementation with cardiovascular disease and stroke in RCTs.

Refining the Detection of Systemic Congestion in Severe Tricuspid Regurgitation: Echocardiography May Be  the Key
A growing number of studies have shown the independent prognostic value of tricuspid regurgitation (TR) and the direct link between TR severity and adverse outcomes.1-4 However, TR not only affects prognosis but has also a negative impact on quality of life,5-7 decreasing functional capacity and increasing systemic congestion.7,8 The systolic blood backflow through the incompetent tricuspid valve reduces the forward stroke volume and cardiac output of the right ventricle (RV). Therefore, as TR worsens, pulmonary perfusion decreases, as does the venous return of oxygenated blood from the pulmonary circulation into the left ...

The TICE Pathway: Mechanisms and Potential Clinical Applications
This article focuses on the research progress of the TICE pathway in the last few years, including the discovery process of the TICE pathway, its molecular mechanism, and potential clinical applications.Recent FindingsCholesterol homeostasis is vital for cardiovascular diseases, stroke, and neurodegenerative diseases. Beyond the cholesterol excretion via hepatobiliary pathway, TICE contributes significantly to reverse cholesterol transport ex vivo and in vivo. Nuclear receptors are ligand-activated transcription factors that regulate cholesterol metabolism. The farnesoid X receptor (FXR) and liver X receptor (LXR) activate...

Oral Anticoagulation Use and Left Atrial Appendage Occlusion in LAAOS III
CONCLUSIONS: The benefit of LAA occlusion was consistent whether patients were receiving OACs or not. LAA occlusion provides thromboembolism reduction in patients independent of OAC use.PMID:37732457 | DOI:10.1161/CIRCULATIONAHA.122.060315

Prevalence of atrial septal defects and patent foramen ovale in a cohort of sudden cardiac death patients undergoing autopsy
CONCLUSIONS: The combined rate of PFO and ASD in a cohort of 517 patients undergoing autopsy was 7.9 %. None of these patients had experienced a cerebrovascular accident. This rate of PFOs appears lower than earlier reports and raises the possibility that the relative risk of an associated stroke could be higher than previously estimated.PMID:37734655 | DOI:10.1016/j.jjcc.2023.09.006

French National Diagnostic and Care Protocol for antiphospholipid syndrome in adults and children
Rev Med Interne. 2023 Sep 19:S0248-8663(23)00711-7. doi: 10.1016/j.revmed.2023.08.004. Online ahead of print.ABSTRACTAntiphospholipid syndrome (APS) is a chronic autoimmune disease involving vascular thrombosis and/or obstetric morbidity and persistent antibodies to phospholipids or certain phospholipid-associated proteins. It is a rare condition in adults and even rarer in children. The diagnosis of APS can be facilitated by the use of classification criteria based on a combination of clinical and biological features. APS may be rapidly progressive with multiple, often synchronous thromboses, resulting in life-threatening...

A cardiovascular polypill for secondary stroke prevention in a tertiary centre in Ghana (SMAART): a phase 2 randomised clinical trial
Lancet Glob Health. 2023 Oct;11(10):e1619-e1628. doi: 10.1016/S2214-109X(23)00347-9.ABSTRACTBACKGROUND: A cardiovascular polypill containing generic drugs might facilitate sustained implementation of and adherence to evidence-based treatments, especially in resource-limited settings. However, the impact of a cardiovascular polypill in mitigating atherosclerotic risk among stroke survivors has not been assessed. We aimed to compare a polypill regimen with usual care on carotid intima-media thickness (CIMT) regression after ischaemic stroke.METHODS: In SMAART, a phase 2 parallel, open-label, assessor-masked, randomised clini...

Noncardiac comorbidities and intensive up ‐titration of oral treatment in patients recently hospitalized for heart failure: insights from the STRONG‐HF trial
ConclusionsIn the STRONG-HF trial, non-cardiac comorbidities neither limited the rapid up-titration of HF-therapies, nor attenuated the benefit of HIC on primary endpoint. In the context of a clinical trial, the benefit-risk ratio favors the rapid up-titration of HF-therapies even in patients with multiple NCCsThis article is protected by copyright. Atherosclerosis as a Risk Factor for IBD: A Population-Based Case-Control Study
CONCLUSIONS: A history of an atherosclerotic-related condition is associated with increased odds of developing IBD, particularly among older adults. Future studies should investigate whether drugs targeting atherosclerotic-related inflammation may prevent IBD in higher-risk individuals.PMID:37721310 | DOI:10.14309/ajg.0000000000002502
