Effect of Inclisiran on Haematological and Immunological Biomarkers: A Pooled Analysis of ORION-9, -10 and -11 Trial Data
Haematological and immunological adverse effects and significant injection-site adverse events (AEs) have been reported with some early generation anti-sense oligonucleotide (ribonucleic acid [RNA]-based) therapies, which required frequent dosing and had non-specific organ uptake. Inclisiran is a novel small interfering RNA (siRNA) therapy with twice-yearly dosing, and conjugated with trivalent N-Acetylgalactosamine (GalNAc) enabling targeted hepatic delivery. Here, we assessed the effects of inclisiran on haematological and immunological biomarkers.

Pooled Safety Analysis of Inclisiran in 3,576 Patients With Approximately 10,000 Person Years of Exposure From Seven Trials
Inclisiran, an siRNA targeting PCSK9 mRNA, provides time-averaged reductions in LDL-C of ∼50% with twice-yearly administration after initial and three-month doses. Pooled data from seven completed or open-label extension trials provide additional data on the long-term safety of inclisiran.
