FXYD3 facilitates Na < sup > + < /sup > and liquid absorption across human airway epithelia by increasing the transport capacity of the Na/K ATPase
Am J Physiol Cell Physiol. 2022 Aug 22. doi: 10.1152/ajpcell.00047.2022. Online ahead of print.ABSTRACTNa/K ATPase activity is essential for ion transport across epithelia. FXYD3, a ᵯE; subunit of the Na/K ATPase, is expressed in the airway, but its function remains undetermined. Single cell RNA sequencing and immunohistochemistry revealed that FXYD3 localizes within the basolateral membrane of all airway epithelial cells. To study FXYD3 function, we reduced FXYD3 expression using siRNA. After permeabilizing the apical membrane with nystatin, epithelia pretreated with FXYD3-targeting siRNA had lower ouabain-sensitive sho...

In vivo antifungal activity and biocompatibility of Cryptocarya moschata
The objective of this study was evaluate, in vivo model, the antifungal activity of Cryptocarya moschata extract against Candida albicans and its biocompatibility. The animals (N = 50) were divided into groups (n = 5): CI/CG: candidiasis was induced and treated with C. moschata extract (0.045 g/mL); CI/NG: candidiasis was induced and treated with nystatin; CI/NT: candidiasis was induced and no treated; CI/CG-2: candidiasis was induced and treated with C. moschata extract (0.045 g/mL), reapplied after 24 h; CI/NG-2: candidiasis was induced and treated with nystatin, reapplied after 24 h; NCI/NT: candidiasis was not induced ...

Natamycin sequesters ergosterol and interferes with substrate transport by the lysine transporter Lyp1 from yeast
Biochim Biophys Acta Biomembr. 2022 Jul 29:184012. doi: 10.1016/j.bbamem.2022.184012. Online ahead of print.ABSTRACTNatamycin is a polyene macrolide, widely employed to treat fungal keratitis and other yeast infections as well as to protect food products against fungal molds. In contrast to other polyene macrolides, such as nystatin or amphotericin B, natamycin does not form pores in yeast membranes, and its mode of action is not well understood. Here, we have employed a variety of spectroscopic methods, computational modeling, and membrane reconstitution to study the molecular interactions of natamycin underlying its anti...

Biotechnological potential of microorganisms from landfill leachate: isolation, antibiotic resistance and leachate discoloration
An Acad Bras Cienc. 2022 Jul 18;94(3):e20210642. doi: 10.1590/0001-3765202220210642. eCollection 2022.ABSTRACTDisposal of municipal solid waste (MSW) can be considered a risk to human health representing a great environmental problem in several countries. MSW landfills are a significant source of toxic elements in the environment. Microorganisms able to thriving in leachate wastewater may exhibit metabolic machinery to synthesize a wide range of enzymes able to degrade and/or discolor toxic compounds from leachate. The use of non-pathogenic microbial cells for human health, recovered from leachate for biotechnological appl...
