Sensitive quantitative analysis of psilocin and psilocybin in hair samples from suspected users and their distribution in seized hallucinogenic mushrooms
ConclusionsThe concentrations of psilocin in two authentic hair were 161 and 150  pg/mg, respectively, and psilocybin was not detected from both samples. This method was also used to analyze the distribution of psilocin and psilocybin in seven hallucinogenic mushrooms. To our knowledge, this is the first demonstration of psilocin concentrations in hair samples of hallucinogenic mushroom consumers, and also our method is most sensitive for quantitative analysis of psilocin and psilocybin in hair samples. A fluorine turns a medicinal benzodiazepine into NPS: the case of flualprazolam
ConclusionsThe recent flualprazolam-related cases have raised concerns to regulatory authorities and international stakeholders suggesting that flualprazolam should be under international control. The urgent international control of flualprazolam, despite the limited information on clinical effects and pharmacologic characteristics available, is an important measure for the prevention of its increasing abuse worldwide. Fatal intoxication with U-47700 in combination with other NPS ( N -ethylhexedrone, adinazolam, 4-CIC, 4-CMC) confirmed by identification and quantification in autopsy specimens and evidences
ConclusionsThe described case of fatal intoxication with NPS presented postmortem concentrations of U-47700, 4-CMC,N-ethylhexedrone, adinazolam and 4-CIC for the first time in the literature. The paper also showed stability study of these substances stored at  + 4 °C for 775 days. (Source: Forensic Toxicology)

Counterfeit formulations: analytical perspective on anorectics
ConclusionsSlimming formulations doped with anorectic cocktails to boost their weight-reducing efficacy are not uncommon. Liquid chromatography combined with mass spectrometry remains the gold standard for counterfeit drug analysis, and requires improved preconcentration methods for rapid and quantitative identification of specific chemical constituents. Extensive method development and validation, targeted at refining existing techniques while developing new ones, is expected to improve the analytical profiling of counterfeit anorectics significantly. Discovering the major metabolites of the three novel fentanyl analogues 3-methylcrotonylfentanyl, furanylbenzylfentanyl, and 4-fluorocyclopropylbenzylfentanyl for forensic case work
ConclusionsIn the present study, we successfully discovered and elucidated the structures of the major metabolites of 3-MCF, FBF, and 4-FCBF which could be used as markers to confirm intake of these compounds in forensic case work. (Source: Forensic Toxicology)

Identification and structure characterization of five synthetic opioids: 3,4-methylenedioxy-U-47700, o -methyl-acetylfentanyl, 2-thiophenefentanyl, benzoylfentanyl and benzoylbenzylfentanyl
ConclusionsComplementary LC –QTOF-MS/MS and NMR techniques enabled the unambiguous identification of new synthetic opioids. To the best of our knowledge, this is the first comprehensive characterization of these five NPS with complete data which are published to facilitate their detection and identification by forensic and c linical laboratories. Monitoring metabolism of synthetic cannabinoid 4F-MDMB-BINACA via high-resolution mass spectrometry assessed in cultured hepatoma cell line, fungus , liver microsomes and confirmed using urine samples
ConclusionsThe in-vitro models presented proved useful in the exhaustive metabolism studies. Despite limitations, HepG2 identified the major 4F-MDMB-BINACA ester hydrolysis metabolite, andC. elegans demonstrated the capacity to produce a wide variety of metabolites. BothC. elegans and HLM produced all the in-vivo metabolites. Ester hydrolysis and ester hydrolysis plus  dehydrogenation 4F-MDMB-BINACA metabolites were recommended as urinary markers for 4F-MDMB-BINACA intake. (Source: Forensic Toxicology)

Development and validation of quantitative analytical method for 50 drugs of antidepressants, benzodiazepines and opioids in oral fluid samples by liquid chromatography –tandem mass spectrometry
ConclusionsThe method was validated according to international recommendations for the 50 analytes, showing low limits of quantification, good imprecision and bias values, using simple liquid –liquid extraction, and was successfully applied to authentic oral fluid samples analysis.
