Development of a 64Cu-labeled CD4+ T cell targeting PET tracer: evaluation of CD4 specificity and its potential use in collagen-induced arthritis
ConclusionsWe developed and evaluated a new radiotracer, [64Cu]Cu-NOTA-CD4, for immunoPET imaging of murine CD4+ T cells. [64Cu]Cu-NOTA-CD4 was successfully synthesized by F(ab) ’2 fragments of R-anti-mouse CD4 antibodies conjugated to a chelator and radiolabeled with copper-64. We found that our novel CD4 PET tracer can be used for noninvasive visualization of murine CD4+ T cells. (Source: EJNMMI Research)

PARP targeted Auger emitter therapy with [125I]PARPi-01 for triple-negative breast cancer
ConclusionConsidering the radio-cytotoxic effect in the tumour tissue and a delay on tumour doubling time, [125I]PARPi-01 presents a potential radiotherapeutics for treatment of TNBC. Improvements to overcome the suboptimal pharmacokinetics are necessary for its potential clinical application. (Source: EJNMMI Research)

Effect of steroid treatment on the diagnostic yield of baseline 18f-fluorodeoxyglucose positron emission tomography in aggressive B cell lymphoma
AbstractAggressive B cell lymphoma often requires prompt steroid treatment, even before baseline18f-fluorodeoxyglucose (FDG) positron emission tomography/computed tomography (PET/CT) and definitive treatment, to alleviate symptoms or prevent organ damage. Since lymphoma is a steroid-sensitive malignancy, there are concerns that steroids might affect the results of FDG PET/CT and decrease its diagnostic yield. The aim of the current study was to evaluate the effect of steroids administered before baseline PET/CT on the maximum standardized uptake value (SUVmax) and additional PET/CT parameters. Retrospective review of the d...

External validation: a simulation study to compare cross-validation versus holdout or external testing to assess the performance of clinical prediction models using PET data from DLBCL patients
ConclusionIn case of small datasets, it is not advisable to use a holdout or a very small external dataset with similar characteristics. A single small testing dataset suffers from a large uncertainty. Therefore, repeated CV using the full training dataset is preferred instead. Our simulations also demonstrated that it is important to consider the impact of differences in patient population between training and test data, which may ask for adjustment or stratification of relevant variables. Short-time-window Patlak imaging using a population-based arterial input function and optimized Bayesian penalized likelihood reconstruction: a feasibility study
ConclusionsShort-time-window Ki images with a population-based IF reconstructed using the BPL algorithm and a high beta value were closely correlated with long-time-window Ki images generated with an individual patient-based IF. Short-time-window Ki images using a population-based IF and BPL reconstruction might represent practical alternatives to long-time-window Ki images generated using an individual patient-based IF. [18F]FE-PE2I PET is a feasible alternative to [123I]FP-CIT SPECT for dopamine transporter imaging in clinically uncertain parkinsonism
ConclusionsThe high correspondence of [18F]FE-PE2I PET compared to reference standard [123I]FP-CIT SPECT establishes [18F]FE-PE2I PET as a feasible PET tracer for clinical use with favourable scan logistics. (Source: EJNMMI Research)

Change in brain amyloid load and cognition in patients with amnestic mild cognitive impairment: a 3-year follow-up study
ConclusionsIn the early phase of aMCI, presence of amyloid pathology at baseline strongly predicted amyloid accumulation during follow-up, which was further paralleled by cognitive declines. Inversely, some of our patients remained amyloid negative also at the end of the study without significant change in18F-Flutemetamol uptake or cognition. Future studies with longer follow-up are needed to distinguish whether the underlying pathophysiology of aMCI in such patients is other than AD. (Source: EJNMMI Research)

Optimal imaging time points considering accuracy and precision of Patlak linearization for 89Zr-immuno-PET: a simulation study
ConclusionInclusion of a blood sample and PET scan at 24  h p.i. improves accuracy and precision of Patlak results for89Zr-immuno-PET; the exact timing of the two later time points is not critical. (Source: EJNMMI Research)

Biodistribution and dosimetry of the GluN2B-specific NMDA receptor PET radioligand (R)-[11C]Me-NB1
ConclusionThe GluN2B-specific radioligand(R)-[11C]Me-NB1 was well-tolerated without reported side effects. Effective dose was estimated to 1.8  mSv when using 300 MBq of presented radioligand. The critical organ was the urinary bladder. Due to the low effective dose coefficient of this radioligand, longitudinal studies for drug development and treatment monitoring of neuropsychiatric disorders including neurodegenerative diseases are pos sible.Trial registration Registered on 11th of June 2019 athttps://www.basg.gv.at (EudraCT: 2018-002933-39). A guideline proposal for mice preparation and care in 18F-FDG PET imaging
AbstractThe experimental outcomes of small-animal positron emission tomography (PET) imaging with18F-labelled fluorodeoxyglucose (18F-FDG) can be particularly compromised by animal preparation and care. Several works intend to improve research reporting and amplify the quality and reliability of published research. Though these works provide valuable information to plan and conduct animal studies, manuscripts describe different methodologies —standardization does not exist. Consequently, the variation in details reported can explain the difference in the experimental results found in the literature. Additionally, the res...
