Evaluation of the Potential for Cytochrome P450 and Transporter-Mediated Drug-Drug Interactions for Cilofexor, a Selective Nonsteroidal Farnesoid X Receptor (FXR) Agonist
ConclusionCilofexor may be coadministered with inhibitors of P-gp, CYP3A4, or CYP2C8 without the need for dose modification. Cilofexor may be coadministered with OATP, BCRP, P-gp, and/or CYP3A4 substrates —including statins—without dose modification. However, coadministration of cilofexor with strong hepatic OATP inhibitors, or with strong or moderate inducers of OATP/CYP2C8, is not recommended. (Source: Clinical Pharmacokinetics)

Comparison of radon adsorption properties under dynamic radon exhalation condition and characteristics of three biochar
AbstractRice husk, grapefruit peel and sugarcane waste residue were used as raw materials to prepare biochar. The radon adsorption performance of biochar under dynamic radon exhalation environment was tested and the characterization of biochar was analyzed. The radon adsorption performance of PC600-D (rice husk biochar) and PC600-G (sugarcane bagasse biochar) was much better than that of PC600-Y (grapefruit peel biochar). The type of pore structure and the percentage of pore area had a greater influence on the biochar, and the honeycomb regular isolation structure of PC600-D and The regular honeycomb isolation structure of...

Severity of coeliac disease and clinical management study when using a non-metabolised medication: a phase I pharmacokinetic study
Conclusions Coeliac disease severity based on duodenal histopathology did not affect oral fexofenadine bioavailability. Increased Tmax suggested absorption distal to the duodenum (jejunum + ileum), where histology seems more normal which may be the key determinant. Patients with coeliac disease may not require consideration for alternative clinical drug management for a number of non-metabolised and transport-mediated medications. (Source: BMJ Open)

Food effects on gastrointestinal physiology and drug absorption
Drug Metab Pharmacokinet. 2022 Dec 20:100488. doi: 10.1016/j.dmpk.2022.100488. Online ahead of print.ABSTRACTFood ingestion affects the oral absorption of many drugs in humans. In this review article, we summarize the physiological factors in the gastrointestinal (GI) tract that affect the in vivo performance of orally administered solid dosage forms in fasted and fed states in humans. In particular, we discuss the effects of food ingestion on fluid characteristics (pH, bile concentration, and volume) in the stomach and small intestine, GI transit of water and dosage forms, and microbiota. Additionally, case examples of fo...
