Safety review of current pharmacotherapies for levodopa-treated patients with Parkinson's disease
Expert Opin Drug Saf. 2023 Jul 4:1-17. doi: 10.1080/14740338.2023.2227096. Online ahead of print.ABSTRACTINTRODUCTION: Levodopa remains the gold standard for treatment of Parkinson's disease (PD). Patients develop complications with disease progression, necessitating adjunctive therapy to control fluctuations in motor and non-motor symptoms and dyskinesia. Knowledge of medication safety and tolerability is critical to ascertain the benefit-risk ratio and select an adjunctive therapy that provides the highest chance for medication adherence. Posing a challenge are the sheer abundance of options, stemming from the developmen...

Adamantyl Amines Activity Study Against Mutant Influenza A M2 Channels Identified a Polycyclic Cage Amine Triple blocker Explored with MD simulations and Solid state NMR
ChemMedChem. 2023 Jun 28:e202300182. doi: 10.1002/cmdc.202300182. Online ahead of print.ABSTRACTWe compared the anti-influenza potencies of 57 adamantyl amines and analogs against influenza A virus with serine-31 M2 proton channel, usually termed as WT M2 channel, which is amantadine sensitive. We also tested a sub-set of these compounds against viruses with the amantadine resistant L26F, V27A, A30T, G34E M2 mutant channels. (a) Four compounds inhibited WT M2 virus in vitro with mid-nanomolar and 27 compounds with sub-micromolar to low micromolar potency. (b) Several compounds inhibited L26F M2 virus in vitro with sub-micr...

Prescribing patterns and determinants for elderly patients with Parkinson's disease in Japan: a retrospective observational study using insurance claims databases
ConclusionPrescribing patterns for patients ≥75 years were levodopa centered and less complex than for those <75 years. Factors significantly associated with levodopa monotherapy and continued use of levodopa were older age and cognitive disorder.Clinical trial registrationUMIN Clinical Trials Registry, https://center6.umin.ac.jp/cgi-open-bin/ctr_e/ctr_view.cgi?recptno=R000053425 (UMIN000046823). Structurally dynamic self-healable hydrogel cooperatively inhibits intestinal inflammation and promotes mucosal repair for enhanced ulcerative colitis treatment
Biomaterials. 2023 May 31;299:122184. doi: 10.1016/j.biomaterials.2023.122184. Online ahead of print.ABSTRACTHydrogels are a class of biocompatible materials with versatile functions that have been increasing explored for the localized treatment of ulcerative colitis (UC), but various mechanical stimuli may cause premature hydrogel breakage and detachment, impeding their further clinical translation. Here we report a multifunctional mechanically-resilient self-healing hydrogel for effective UC treatment, which is synthesized through the host-guest interaction between dopamine/β-cyclodextrin-modified hyaluronic acid (HA-CD...
