Combinatorial treatment of idiopathic pulmonary fibrosis using nanoparticles with prostaglandin E and siRNA(s)
Publication date: Available online 19 April 2017 Source:Nanomedicine: Nanotechnology, Biology and Medicine Author(s): O.B. Garbuzenko, V. Ivanova, V. Kholodovych, D.C. Reimer, K.R. Reuhl, E. Yurkow, D. Adler, T. Minko Inhalation delivery of prostaglandin E (PGE2) in combination with selected siRNA(s) was proposed for the efficient treatment of idiopathic pulmonary fibrosis (IPF). Nanostructured lipid carriers (NLC) were used as a delivery system for PGE2 with and without siRNAs targeted to MMP3, CCL12, and HIF1Alpha mRNAs. The model of IPF was developed in SKH1 mice by intratracheal administration of bleomycin at a dose o...

RNA Delivery: Modularized Gold Nanocarriers for TAT ‐Mediated Delivery of siRNA (Small 8/2017)
A novel plasmonic gold nanocarrier using a modular RNA scaffold significantly improves delivery efficiency into diverse human cells, as presented by Norbert O. Reich and co‐workers in article number 1602473. The orthogonal positioning of cell targeting peptides and functional RNA provides unprecedented control in the delivery of biologically active RNA. NIR light‐triggered RNA release with spatio‐temporal control further enhances the RNA delivery efficiency.

Pancreatic Cancer: Targeted Co ‐delivery of PTX and TR3 siRNA by PTP Peptide Modified Dendrimer for the Treatment of Pancreatic Cancer (Small 2/2017)
A new type of tumor‐targeted nanovehicle (PSPGP) is successfully synthesized for co‐delivery of PTX (paclitaxel) and TR3 siRNA, by G. Yu, G. Tang, and co‐workers in article number 1602697. In vitro and in vivo investigations demonstrate that the redox‐responsive PSPGP exhibited enhanced endosomal escape and intracellular degradation, which facilitates PTX and TR3 siRNA release, effectively improving the anti‐tumor efficacy.

Design of lipid nanoparticles for in vitro and in vivo delivery of plasmid DNA
Publication date: Available online 28 December 2016 Source:Nanomedicine: Nanotechnology, Biology and Medicine Author(s): Jayesh A. Kulkarni, J. Layne Myhre, Sam Chen, Yuen Yi C. Tam, Adrian Danescu, Joy M. Richman, Pieter R. Cullis Lipid nanoparticles (LNPs) containing distearolyphosphatidlycholine (DSPC), and ionizable amino-lipids such as (6Z,9Z,28Z,31Z)-Heptatriaconta-6,9,28,31-tetraen-19-yl-4-(dimethylamino)butanoate (DLin-MC3-DMA) are potent siRNA delivery vehicles in vivo. Here we explore the utility of similar LNP systems as transfection reagents for plasmid DNA (pDNA). It is shown that replacement of DSPC by unsat...

Nanoparticles for siRNA-Based Gene Silencing in Tumor Therapy
Gene silencing through RNA interference (RNAi) has emerged as a potential strategy in manipulating cancer causing genes by complementary base-pairing mechanism. Small interfering RNA (siRNA) is an important RNAi tool that has found significant application in cancer therapy. However due to lack of stability, poor cellular uptake and high probability of loss-of-function due to degradation, siRNA therapeutic strategies seek safe and efficient delivery vehicles for in vivo applications. The current review discusses various nanoparticle systems currently used for siRNA delivery for cancer therapy, with emphasis on liposome base...

Nanoparticle delivery of siRNA against TWIST to reduce drug resistance and tumor growth in ovarian cancer models
Publication date: Available online 25 November 2016 Source:Nanomedicine: Nanotechnology, Biology and Medicine Author(s): Cai M. Roberts, Sophia Allaf Shahin, Wei Wen, James B. Finlay, Juyao Dong, Ruining Wang, Thanh H. Dellinger, Jeffrey I. Zink, Fuyuhiko Tamanoi, Carlotta A. Glackin Epithelial ovarian cancer (EOC) is the most deadly gynecologic malignancy on account of its late stage at diagnosis and frequency of drug resistant recurrences. Novel therapies to overcome these barriers are urgently needed. TWIST is a developmental transcription factor reactivated in cancers and linked to angiogenesis, metastasis, cancer ste...

Modularized Gold Nanocarriers for TAT ‐Mediated Delivery of siRNA
Targeted delivery of siRNA controlled by near‐infrared light using hollow gold nanoshells has been demonstrated in cancer and stem cells models. Here, a universal surface module and several functionalization rules for the maximized delivery of short nucleic acids (here, siRNA) applicable for diverse gold nanocarriers are described. Streptavidin is devised as a handle to assemble biotinylated cell penetrating peptides (e.g., transactivating transcriptional activator (TAT)), as well as an insulator between the positive charge of TAT and the dense negative charge of RNA. However, direct linking of streptavidin to functional...

Hollow Carbon Nanospheres with Tunable Hierarchical Pores for Drug, Gene, and Photothermal Synergistic Treatment
Design and synthesis of porous and hollow carbon spheres have attracted considerable interest in the past decade due to their superior physicochemical properties and widespread applications. However, it is still a big challenge to achieve controllable synthesis of hollow carbon nanospheres with center‐radial large mesopores in the shells and inner surface roughness. Herein, porous hollow carbon nanospheres (PHCNs) are successfully synthesized with tunable center‐radial mesopore channels in the shells and crater‐like inner surfaces by employing dendrimer‐like mesoporous silica nanospheres (DMSNs) as hard templates. ...
