Peptidyl Virus ‐Like Nanovesicles as Reconfigurable “Trojan Horse” for Targeted siRNA Delivery and Synergistic Inhibition of Cancer Cells
Peptidyl virus-like nanovesicles (pVLNs) composed of bilayer membranes are assembled by the peptides and siRNA. The targeting and enzyme-responsive sequences on the bilayer's surface allow the pVLNs to enter cancer cells with high efficiency and control the release of genetic drugs in response to the subcellular environment. The resulting gene silencing efficiency is 92% and cancer cell mortality is 96%. AbstractThe self-assembly of peptidyl virus-like nanovesicles (pVLNs) composed of highly ordered peptide bilayer membranes that encapsulate the small interfering RNA (siRNA) is reported. The targeting and enzyme-responsive...

TfR Aptamer Enhanced Blood ‐Brain Barrier Penetration of Biomimetic Nanocomplexes for Intracellular Transglutaminase 2 Imaging and Silencing in Glioma
Biomimetic nanocomplex (TMPsM) nanopartcles with dual blood-brain barrier (BBB)  targeting capability are prepared for simultaneous accurate diagnosis and effective treatment of glioma through single intracellular transglutaminase 2-triggered self-assembly aggregation-induced emission imaging and RNAi therapy. The dual BBB targeting biomimetic outer shells endow it with enhanc ed BBB penetration, prolonged blood circulation, and ensure the delivery of inner cores into brain tumor sites for theranostic agents release. AbstractEngineering a versatile nanocomplex integrating effective penetration of the blood-brain barrier (...

Unprotonatable and ROS ‐Sensitive Nanocarrier for NIR Spatially Activated siRNA Therapy with Synergistic Drug Effect
An unprotonatable and reactive oxygen species-responsive nanocarrier is designed to co-deliver photodynamic reagent chlorin e6 (Ce6) and small interfering RNA (siRNA). Owing to the unprotonatable property and near infrared-activated photochemical internalization effect, the cationic nanocarrier spatially controls siRNA activity “on/off”, turning on siRNA therapy with the synergistic effect of Ce6-based photodynamic therapy in tumor tissue and turning off siRNA activity in normal tissues. AbstractAlthough small interfering RNA (siRNA) therapy has achieved great progress, unwanted gene inhibition in normal tissues severe...
