Delivery of siRNA using folate receptor-targeted pH-sensitive polymeric nanoparticles for rheumatoid arthritis therapy
Publication date: Available online 22 May 2019Source: Nanomedicine: Nanotechnology, Biology and MedicineAuthor(s): Xiangshi Sun, Shiyan Dong, Xiangyu Li, Kongtong Yu, Fengying Sun, Robert J. Lee, Youxin Li, Lesheng TengAbstractSystemic delivery of siRNA to target tissues is difficult to achieve owing to its limited cellular uptake and poor serum stability. Herein, polymeric nanoparticles were developed for systemic administration of siRNA to inflamed tissues. The polymeric nanoparticles were composed of PK3 as a pH-sensitive polymer, folate-polyethyleneglycol-poly(lactide-co-glycolide) as a targeting ligand, and a DOTAP/si...

Silencing AXL by covalent siRNA-gelatin-antibody nanoconjugate inactivates mTOR/EMT pathway and stimulates p53 for TKI sensitization in NSCLC
Publication date: Available online 12 May 2019Source: Nanomedicine: Nanotechnology, Biology and MedicineAuthor(s): Dhananjay Suresh, Ajit Zambre, Soumavo Mukherjee, Shreya Ghoshdastidar, Yuexu Jiang, Trupti Joshi, Anandhi Upendran, Raghuraman KannanAbstractNon-small cell lung cancer (NSCLC) is the leading cause of cancer mortality with the 5-year survival rate remains a dismal 16% for the past 40 years. Drug resistance is a major obstacle to achieving long-term patient survival. Identifying and validating molecular biomarkers responsible for resistance and thereby adopting multi-directional therapy is necessary to imp...

Sequential Targeting TGF ‐β Signaling and KRAS Mutation Increases Therapeutic Efficacy in Pancreatic Cancer
Fraxinellone ‐loaded CGKRK‐modified nanoparticles are prepared for regulating transforming growth factor beta signals in the suppressive tumor microenvironment. Based on this, the dense stroma is modulated and the intratumor blood perfusion is enhanced, meaning the delivery barrier of the following siRNA ‐loaded lipid‐coated calcium phosphate (LCP) biomimetic high‐density lipoprotein nanoparticles (siKras‐LCP‐ApoE3) are internalized and specifically silence KRAS mutation and kill cancer cells. AbstractPancreatic cancer is a highly aggressive malignancy that strongly resists extant treatments. The failure of e...

Cationic lipoplexes for treatment of cancer stem cell-derived murine lung tumors
This study outlines the fabrication of a lipoplex of SOX2 small interfering RNA (CL-siSOX2) for targeted treatment of SOX2-enriched, CSC-derived orthotopic and xenograft lung tumors in CB-17 SCID mice. CL-siSOX2 induced tumor contraction in cisplatin-naïve and cisplatin-treated groups by 85% and 94% respectively. Reduction in tumor weight and volume following treatment with CL-siSOX2 was associated with reduced protein expression of SOX2 and markers of tumor initiation, inflammation, invasion and metastasis in mice tumor xenografts. In addition, histological staining of lung tumor sections showed reduction in SOX2 express...

Chronic administration of nano-sized PAMAM dendrimers in vivo inhibits EGFR-ERK1/2-ROCK signaling pathway and attenuate diabetes-induced vascular remodeling and dysfunction
This study showed chronic in vivo administration of PAMAM dendrimers in an animal model of diabetes inhibited EGFR-ERK1/2-ROCK signaling and improved diabetes-induced vascular remodeling and dysfunction in a dose, generation (G6 > G5) and surface chemistry-dependent manner (cationic> anionic> neutral). Thus, naked PAMAM dendrimers have the propensity to modulate key (e.g. EGFR) cell signaling cascades with associated pharmacological consequences in vivo that are dependent on their physicochemical properties. PAMAMs, alone or in combination with vasculoprotective agents, might therefore have a beneficial role in t...
