Improving the Enantioselectivity of CHMO Brevi1  for Asymmetric Synthesis of Podophyllotoxin Precursor
In this study, we use a strategy called "focused rational iterative site-specific mutagenesis" (FRISM) at residues ranging of 6 Å from substrate. The mutations by using a restricted set of rationally chosen amino acids allows the formation of a small mutant library. By generating and screening less than 60 variants, we achieved high ee of 96.8%. Coupled with the cofactor regeneration system, 9.3 mM substrate was converted completely in a 100-mL scale reaction. Therefore, our work reveals a promising synthetic method for (R)-β-piperonyl-γ-butyrolactone with the highest enantioselectivity, and provides a new opportunity f...

Critical functions of extracellular matrix in brain metastasis seeding
Cell Mol Life Sci. 2023 Sep 20;80(10):297. doi: 10.1007/s00018-023-04944-z.ABSTRACTHuman brain is characterized by extremely sparse extracellular matrix (ECM). Despite its low abundance, the significance of brain ECM in both physiological and pathological conditions should not be underestimated. Brain metastasis is a serious complication of cancer, and recent findings highlighted the contribution of ECM in brain metastasis development. In this review, we provide a comprehensive outlook on how ECM proteins promote brain metastasis seeding. In particular, we discuss (1) disruption of the blood-brain barrier in brain metastas...

Single-cell genotypic and phenotypic analysis of measurable residual disease in acute myeloid leukemia
Sci Adv. 2023 Sep 22;9(38):eadg0488. doi: 10.1126/sciadv.adg0488. Epub 2023 Sep 20.ABSTRACTMeasurable residual disease (MRD), defined as the population of cancer cells that persist following therapy, serves as the critical reservoir for disease relapse in acute myeloid leukemia and other malignancies. Understanding the biology enabling MRD clones to resist therapy is necessary to guide the development of more effective curative treatments. Discriminating between residual leukemic clones, preleukemic clones, and normal precursors remains a challenge with current MRD tools. Here, we developed a single-cell MRD (scMRD) assay ...

Shuttle between arginine and lysine: influence on cancer immunonutrition
Amino Acids. 2023 Sep 20. doi: 10.1007/s00726-023-03327-9. Online ahead of print.ABSTRACTAmino acids which are essential nutrients for all cell types' survival are also recognised to serve as opportunistic/alternative fuels in cancers auxotrophic for specific amino acids. Accordingly, restriction of amino acids has been utilised as a therapeutic strategy in these cancers. Contrastingly, amino acid deficiencies in cancer are found to greatly impair immune functions, increasing mortality and morbidity rates. Dietary and supplemental amino acids in such conditions have revealed their importance as 'immunonutrients' by modulat...

Implications of ubiquitination and the maintenance of replication fork stability in cancer therapy
Biosci Rep. 2023 Sep 20:BSR20222591. doi: 10.1042/BSR20222591. Online ahead of print.ABSTRACTDNA replication forks are subject to intricate surveillance and strict regulation by sophisticated cellular machinery. Such close regulation is necessary to ensure the accurate duplication of genetic information and to tackle the diverse endogenous and exogenous stresses that impede this process. Stalled replication forks are vulnerable to collapse, which is a major cause of genomic instability and carcinogenesis. Replication stress responses, which are organized via a series of coordinated molecular events, stabilize stalled repli...

CRISPR-mediated reversion of oncogenic KRAS mutation results in increased proliferation and reveals independent roles of Ras and mTORC2 in the migration of A549 lung cancer cells
Mol Biol Cell. 2023 Sep 20:mbcE23050152. doi: 10.1091/mbc.E23-05-0152. Online ahead of print.ABSTRACTAlthough the RAS oncogene has been extensively studied, new aspects concerning its role and regulation in normal biology and cancer continue to be discovered. Recently, others and we have shown that the mechanistic Target of Rapamycin Complex 2 (mTORC2) is a Ras effector in Dictyostelium and mammalian cells. mTORC2 plays evolutionarily conserved roles in cell survival and migration and has been linked to tumorigenesis. Since RAS is often mutated in lung cancer, we investigated whether a Ras-mTORC2 pathway contributes to enh...
