The Nedd8 ‐activating enzyme inhibitor MLN4924 (TAK‐924/Pevonedistat) induces apoptosis via c‐Myc‐Noxa axis in head and neck squamous cell carcinoma
ConclusionsThese findings determined the over ‐expression levels of neddylation enzymes in HNSCC and revealed novel mechanisms underlying neddylation inhibition induced growth suppression in HNSCC cells, which provided preclinical evidence for further clinical evaluation of neddylation inhibitors (eg, MLN4924) for the treatment of HNSCC.

Constitutive activation and overexpression of NF- κB/c-Rel in conjunction with p50 contribute to aggressive tongue tumorigenesis.
In conclusion, selective participation of c-Rel with p50 that in cross-talk with AP-1/Fra-2 induced poor differentiation and aggressive tumorigenesis mainly in HPV-ve smokers while HPV infection induced expression of p65 and p27 leading to well differentiation and better prognosis preferably in non-smoking TSCC patients. Protein phosphatase 2A (PP2A) inhibitor CIP2A indicates resistance to radiotherapy in rectal cancer
In conclusion, these results suggest that by contributing to radiosensitivity of cancer cells, low CIP2A protein expression level associates with a favorable response to long‐course (C)RT in rectal cancer patients. Low CIP2A protein expression level associates with a favorable response to long‐course chemoradiotherapy in rectal cancer patients. Suppression of CIP2A transcription by siRNA sensitizes colorectal cancer cells to irradiation and decreases their survival in vitro.

10 Knockdown of transforming growth factor beta regulator 4 (tbrg4) promotes apoptosis in human lung cancer cell line h1299 via regulating ddit3, cav1 and rrm2
AbstractTransforming growth factor beta regulator 4 (TBRG4) gene is located on 7 p14-p13, a region correlated with many cancer types, such as hematologic malignancies, head and neck squamous cell carcinoma, and multiple myeloma. However, the role of TBRG4 in human lung cancer is largely unknown. Thus, in the present study, the TBRG4 mRNA level was evaluated in the lung cancer cell line H1299 by quantitative PCR (qPCR) after knocking down TBRG4 using lentivirus-mediated small interfering RNA (siRNA). The result showed that TBRG4 was significantly inhibited in H1299 by RNAi. Microarray analysis revealed 586 differentially-ex...

Minichromosome maintenance (MCM) protein 4 overexpression is a potential prognostic marker for laryngeal squamous cell carcinoma.
CONCLUSIONS: We noticed a significant role for MCM4 overexpression in human LSCC tissues and their corresponding adjacent non-neoplastic tissues and found that siRNA of MCM4 can significantly decrease the proliferation of cancer cells. It is suggested that MCM4 profiling could potentially be used to predict response to treatment and prognosis in LSCC. PMID: 29135113 [PubMed - in process]

Passenger strand of miR-145-3p acts as a tumor-suppressor by targeting MYO1B in head and neck squamous cell carcinoma.
Abstract Analysis of the microRNA (miRNA) expression signature of head and neck squamous cell carcinoma (HNSCC) based on RNA sequencing showed that dual strands of pre‑miR‑145 (miR‑145‑5p, guide strand; and miR‑145‑3p, passenger strand) were significantly reduced in cancer tissues. In miRNA biogenesis, passenger strands of miRNAs are degraded and have no biological activities in cells. The aims of this study were to investigate the functional significance of the passenger strand of miR‑145 and to identify miR‑145‑3p‑regulated oncogenic genes in HNSCC cells. Expression levels of miR‑145‑5p ...

Multilayered omics-based analysis of a head and neck cancer model of cisplatin resistance reveals intratumoral heterogeneity and treatment-induced clonal selection.
CONCLUSIONS: Our study demonstrates a link of intratumoral heterogeneity and clonal evolution as important mechanisms of drug resistance in SCCHN and establishes mutant GOF TP53 variants and the PI3K/mTOR pathway as molecular targets for treatment optimization. DKK3 Overexpression Increases Malignant Properties of Head and Neck Squamous Cell Carcinoma Cells.
Authors: Katase N, Nishimatsu SI, Yamauchi A, Yamamura M, Terada K, Itadani M, Okada N, Hassan NMM, Nagatsuka H, Ikeda T, Nohno T, Fujita S Abstract DKK3, a member of dickkopf WNT signaling pathway inhibitor family, is believed to be a tumor suppressor, because of itsreduced expression in cancer cells. However, our previous studies have revealed that DKK3 expression is predominantly observed in head and neck, and oral squamous cell carcinoma (HNSCC/OSCC). Interestingly, HNSCC/OSCC patients with DKK3 expression showed high metastasis rate and poorer survival, and siRNA-mediated knockdown of DKK3 in HNSCC-derived can...

Livin enhances chemoresistance in head and neck squamous cell carcinoma.
In conclusion, our results suggest that siRNA-mediated Livin knockdown enhanced the chemosensitivity of the three HNSCC cell lines to cisplatin, 5-FU and docetaxel. Although further investigations are required to support these findings, our results demonstrated that novel therapeutic strategies with combined use of siRNA targeting Livin and chemotherapeutic agents may have applications in the treatment of advanced HNSCC. In Vitro and In Vivo Synergistic Antitumor Activity of the Combination of BKM120 and Erlotinib in Head and Neck Cancer: Mechanism of Apoptosis and Resistance
This study investigated whether cotargeting of EGFR and PI3K has synergistic antitumor effects and apoptosis induction. We examined growth suppression, apoptosis, and signaling pathway modulation resulting from single and combined targeting of EGFR and PI3K with erlotinib and BKM120, respectively, in a panel of SCCHN cell lines and a xenograft model of SCCHN. In a panel of 12 cell lines, single targeting of EGFR with erlotinib or PI3K with BKM120 suppressed cellular growth without inducing significant apoptosis. Cotargeting of EGFR and PI3K synergistically inhibited SCCHN cell line and xenograft tumor growth, but induced v...

The Expression Levels of XLF and Mutant P53 Are Inversely Correlated in Head and Neck Cancer Cells
In this study, we have found that XLF is over-expressed in HPV(+) versus HPV(-) head and neck squamous cell carcinoma (HNSCC) and significantly down-regulated in the HNSCC cell lines expressing high level of mutant p53 protein versus those cell lines harboring wild-type TP53 gene with low p53 protein expression. We have also demonstrated that Werner syndrome protein (WRN), a member of the NHEJ repair pathway, binds to both mutant p53 protein and NHEJ1 gene promoter, and siRNA knockdown of WRN leads to the inhibition of XLF expression in the HNSCC cells. Collectively, these findings suggest that WRN and p53 are involved in ...
