Development and validation of clinically Mimicable model of frostbite injury-induced chronic pain
In this study, we employed deep-frozen magnets to induce frostbite injury and conducted validation for chronic pain through assessments of face, predictive, and mechanistic validity. Behavioral assays demonstrated that animals with frostbite injury exhibited significant mechanical, thermal & cold hypersensitivity in injured rats. Further, molecular analysis indicated that frostbite injury triggered the activation of TRP channels (TRPA1 and TRPM8), microgliosis, and increased neuroinflammation in the dorsal root ganglion (DRG) and spinal cord of rats. Notably, NR2B protein expressions were significantly upregulated in t...

LncRNA-HOXC-AS2 regulates tumor-associated macrophage polarization through the STAT1/SOCS1 and STAT1/CIITA pathways to promote the progression of non-small cell lung cancer
In conclusion, HOXC-AS2 inhibited the activation of STAT1/SOCS1 and STAT1/CIITA pathways and promoted M2 polarization of TAMs by binding with STAT1, thus promoting NSCLC.PMID:38168631 | DOI:10.1016/j.cellsig.2023.111031 (Source: Cellular Signalling)

The roles and mechanisms of TGFB1 in acute myeloid leukemia chemoresistance
This study concludes that TGFB1 level in R/R AML patients and drug-resistant strains is significantly increased. Blocking the TGFB signaling pathway can enhance the chemosensitivity of drug-resistant cells by suppressing SOX4 expression and metabolic reprogramming.PMID:38171389 | DOI:10.1016/j.cellsig.2023.111027 (Source: Cellular Signalling)

Enhancer of zeste homolog 2 (EZH2)-dependent sirtuin-3 determines sensitivity to glucose starvation in radioresistant head and neck cancer cells
In this study, we aim to investigate how SIRT3 determines the susceptibility to glucose deprivation and its regulation in p53-based radioresistant head and neck cancer cells. We observed mitochondrial function using two established isogenic radioresistant subclones (HN3R-A [p53 null] and HN3R-B [p53 R282W]) with intratumoral p53 heterogeneity. Cell counting analysis was performed to evaluate cell proliferation and cell death. The correlation between the regulation of SIRT3 and enhancer of zeste homolog 2 (EZH2) was confirmed by immunoblotting and chromatin immunoprecipitation assay. p53-deficient radioresistant cells (HN3R...

GSK3 β phosphorylates Six1 transcription factor and regulates its APC/C < sup > Cdh1 < /sup > mediated proteosomal degradation
Cell Signal. 2023 Dec 30;115:111030. doi: 10.1016/j.cellsig.2023.111030. Online ahead of print.ABSTRACTSine oculis homeobox homolog 1 (Six1) is a developmentally important transcription factor that regulates cellular proliferation, apoptosis, and dissemination during embryogenesis. Six1 overexpression as reported in multiple cancers modulates expression of a repertoire of its target genes causing an increase in proliferation, metastasis and survival of cancer cells. Six1 exists as a cell cycle regulated nuclear phosphoprotein and its cellular turnover is regulated by APC/C (Anaphase promoting complex / Cyclosome) complex m...

A comprehensive evaluation of circ_0065214/ miR-188-3p/GPNMB axis in breast cancer
Cell Signal. 2023 Dec 21;115:111019. doi: 10.1016/j.cellsig.2023.111019. Online ahead of print.ABSTRACTCircRNAs are involved in multiple aspects during carcinogenesis, including tumorigenesis, vascularization, apoptosis and others. Exploring the role of circRNAs in breast cancer (BC) enables us to understand the development mechanism of BC more comprehensively. Here, we screened out and verified an up-regulated circRNA in BC from GEO data. Quantitative Real-time PCR (qRT-PCR) showed that circ_0065214 had a high expression level in BC patients. Besides, circ_0065214 had good diagnostic value in BC serum, and the area under ...
