Hepatitis B virus ‐specific human stem cell memory T cells differentiate into cytotoxic T cells and eradicate HBV‐infected hepatocytes in mice
Here, we detected stem cell memory T cells (TSCMs) in chronic hepatitis B virus (HBV)-infected patients, using HBV core and polymerase peptide HLA tetramers. When these TSCMs were transferred into the human hepatocyte-transplanted, HBV-infected TK-NOG mouse, they differentiated into cytotoxic T lymphocytes, produced interferon gamma and interleukin-2, and developed histologically proven hepatitis. Furthermore, HBV DNA and human albumin in mouse serum declined and human hepatocytes were eliminated in the mouse model. Chronic infection with the hepatitis B virus (HBV) induces progressive hepatic impairment. Achieving complet...

Mathematical Models of Early Hepatitis B Virus Dynamics in Humanized Mice
Bull Math Biol. 2024 Apr 9;86(5):53. doi: 10.1007/s11538-024-01284-2.ABSTRACTAnalyzing the impact of the adaptive immune response during acute hepatitis B virus (HBV) infection is essential for understanding disease progression and control. Here we developed mathematical models of HBV infection which either lack terms for adaptive immune responses, or assume adaptive immune responses in the form of cytolytic immune killing, non-cytolytic immune cure, or non-cytolytic-mediated block of viral production. We validated the model that does not include immune responses against temporal serum hepatitis B DNA (sHBV) and temporal s...

Childhood vaccination coverage and regional differences in Swiss birth cohorts 2012-2021: Are we on track?
CONCLUSION: Full vaccination coverage in Swiss children is still below 90 % with many vaccinations administered delayed. Given regional differences, missed or delayed booster vaccinations, and differences in vaccine-specific acceptability, more effort may be needed to achieve national vaccination targets.PMID:38593195 | DOI:10.1016/j.vaccine.2023.10.043 (Source: Vaccine)

Laboratory diagnostics for vasculitis beyond antineutrophil cytoplasmatic autoantibodies
Z Rheumatol. 2024 Apr 8. doi: 10.1007/s00393-024-01494-y. Online ahead of print.ABSTRACTThe diagnosis of systemic vasculitis (SV) is a major clinical challenge due to the very different forms of presentation and requires an interdisciplinary approach. Targeted laboratory diagnostics support making the diagnosis, differential diagnosis and classification and are also a key component in the detection of active organ manifestations and treatment complications. The basic laboratory tests include the erythrocyte sedimentation rate (ESR), C‑reactive protein (CRP), blood count, serum creatinine, urinalysis, specific autoantibod...

Inhibition of GLUD1 mediated by LASP1 and SYVN1 contributes to hepatitis B virus X protein-induced hepatocarcinogenesis
J Mol Cell Biol. 2024 Apr 8:mjae014. doi: 10.1093/jmcb/mjae014. Online ahead of print.ABSTRACTGlutamate dehydrogenase 1 (GLUD1) is implicated in oncogenesis. However, little is known about the relationship between GLUD1 and hepatocellular carcinoma (HCC). In the present study, we demonstrated that the expression levels of GLUD1 significantly decreased in tumors, which was relevant to the poor prognosis of HCC. Functionally, GLUD1 silencing enhanced the growth and migration of HCC cells. Mechanistically, the upregulation of interleukin-32 through AKT activation contributes to GLUD1 silencing-facilitated hepatocarcinogenesis...
