Wnt Signaling Inhibition Prevents Postnatal Inflammation and Disease Progression in Mouse Congenital Myxomatous Valve Disease
CONCLUSIONS: Wnt signaling is involved in the initiation of mitral valve abnormalities and inflammation but is not responsible for later-stage valve disease progression once it has been initiated. Thus, Wnt signaling contributes to MVD progression in a time-dependent manner and provides a promising therapeutic target for the early treatment of congenital MVD in Marfan syndrome.PMID:38660802 | DOI:10.1161/ATVBAHA.123.320388 (Source: Arteriosclerosis, Thrombosis and Vascular Biology)

Effect of Antimicrobial Agent Coating on Physicochemical and Biologic Properties of Implant Abutments: A Systematic Review
CONCLUSIONS: This review showed that there is still no consensus on which is the better antimicrobial agent and which coated materials have the better performance. However, the association of surface coating of abutments with antimicrobials is feasible and can benefit many patients, which can support their clinical use to favor the healing process and prevent infections that can lead to treatment failure with dental implants.PMID:38657216 | DOI:10.11607/jomi.9992 (Source: International Journal of Oral and Maxillofacial Implants)

High cure rates of Mycoplasma genitalium following empiric treatment with azithromycin alongside frequent detection of macrolide resistance in Austria
ConclusionsMycoplasma genitalium was readily detected in this Austrian observational study, affected predominantly MSM and often presented as asymptomatic disease. We observed a worryingly high prevalence of AZM resistance mutations; however, empiric AZM treatment cleared twice as many MG infections as expected. (Source: Infection)

Leptospira-specific immunoglobulin Y (IgY) is protective in infected hamsters
In conclusion, passive immunotherapy with specific IgY can be considered an effective treatment for leptospirosis, and may replace antibiotics regarding its therapeutic effects.PMID:38641497 | DOI:10.1016/j.vaccine.2024.04.010 (Source: Vaccine)

Case of Haemophilus influenzae urethritis in a male patient
We report a case of H. influenzae urethritis and describe a relatively novel treatment approach. A man presented to Melbourne Sexual Health Centre with a 3-day history of penile discharge and dysuria. He engaged in condomless oral and anal sex with casual male partners in preceding months. On examination, purulent urethral discharge was noted. Urethral microscopy showed Gram-positive bacilli and epithelial cells. Screening for chlamydia, gonorrhoea, Mycoplasma genitalium, Trichomonas vaginalis, HIV and syphilis was performed. He was treated for NGU with a 1-week course of doxycycline. Initial investigations were all negati...

Challenges in the treatment of pediatric Mycoplasma pneumoniae pneumonia
Conclusion:  This report summarizes the clinical significance of macrolide-resistant and refractory MP pneumonia and discusses the efficacy and safety of alternative drugs, with a stepwise approach to the management of MP pneumonia recommended from the viewpoint of clinical practice. What is Known:•Although MP pneumonia is usually a benign self-limited infection with response macrolides as first line therapy, severe life-threatening cases may develop if additional treatment strategies are not effectively implemented.•Macrolide-resistant and refractory MP pneumonia are two conditions that may complicate the clinical cour...

Directed Differentiation of Neurons from Human iPSCs for Modeling Neurological Disorders
Methods Mol Biol. 2024;2794:141-155. doi: 10.1007/978-1-0716-3810-1_12.ABSTRACTHuman-induced pluripotent stem cell (hiPSC) technology has enabled comprehensive human cell-based disease modeling in vitro. Due to limited accessibility of primary human neurons as well as species-specific divergence between human and rodent brain tissues, hiPSC-derived neurons have become a popular tool for studying neuronal biology in a dish. Here, we provide methods for transcription factor-driven directed differentiation of neurons from hiPSCs via a neural progenitor cell (NPC) intermediate. Doxycycline-inducible expression of neuron fate-d...

Peripheral ulcerative keratitis associated with tralokinumab therapy: a case report and review of IL-13 inhibitor-associated ocular surface disease
ConclusionThis case report documents ulcerative keratitis occurring in association with tralokinumab therapy for AD, which has previously been described with dupilumab but not tralokinumab. This report demonstrates the need for vigilant ocular surface monitoring for patients on tralokinumab and illustrates successful management and long-term control of adverse ocular events associated with this medication. Efficacy and safety of antibiotics for treatment of leptospirosis: a systematic review and network meta-analysis
ConclusionsFindings recommend that leptospirosis patients be treated with antibiotics, which significantly reduced the leptospirosis defervescence time. Cephalosporins, doxycycline, and penicillin are suggested, and azithromycin may be a suitable alternative for drug-resistant cases.Systematic review registrationPROSPERO CRD42022354938. Antibiotics Not Associated with Shorter Duration or Reduced Severity of Acute Lower Respiratory Tract Infection
ConclusionsIn this large prospective study in the US primary and urgent care setting, antibiotics had no measurable impact on the severity or duration of cough due to acute LRTI. Patients had unrealistic expectations regarding the duration of LRTI and the effect of antibiotics which should be the target of antibiotic stewardship efforts.
