Premenstrual Dysphoric Disorder
Premenstrual dysphoric disorder (PMDD) comprises emotional and physical symptoms and functional impairment that lie on the severe end of the continuum of premenstrual symptoms. Women with PMDD have a differential response to normal hormonal fluctuations. This susceptibility may involve the serotonin system, altered sensitivity of the GABAA receptor to the neurosteroid allopregnanalone, and altered brain circuitry involving emotional and cognitive functions. Serotonin reuptake inhibitors are considered the first-line treatment. Second-line treatments include oral contraceptives containing drospirenone, other ovulation suppr...

Ethnomedicinal uses of plants for the treatment of malaria in Soon Valley, Khushab, Pakistan
Conclusions To the best of our knowledge seven plant species, viz. Withania coagulans, Fagonia cretica, Carthamus oxyacantha, Ehretia obtusifolia, Helianthus annuus, Olea ferruginea and Vitex trifolia, are reported from this region for the first time for the treatment of malaria. This first ethno-medicinal study highlights potential sources for the development of new antimalarial drugs from indigenous knowledge of medicinal plants found in the Soon Valley, Pakistan. Such investigations could be a subject for in vitro and in vivo anti-plasmodial screening to develop new plant-based antimalarial drugs and can also be evaluat...

Nanoemulsion for improving solubility and permeability of Vitex agnus-castus extract: formulation and in vitro evaluation using PAMPA and Caco-2 approaches.
Authors: Piazzini V, Monteforte E, Luceri C, Bigagli E, Bilia AR, Bergonzi MC Abstract The purpose of this study was to develop new formulation for an improved oral delivery of Vitex agnus-castus (VAC) extract. After the optimization and validation of analytical method for quali-quantitative characterization of extract, nanoemulsion (NE) was selected as lipid-based nanocarrier. The composition of extract-loaded NE resulted in triacetin as oil phase, labrasol as surfactant, cremophor EL as co-surfactant and water. NE contains until 60 mg/mL of extract. It was characterized by DLS and TEM analyses and its ...

A Novel Clerodane Diterpene from Vitex cofassus.
Abstract New clerodane diterpene, 16-hydroxy-pentandralactone (1) and known diterpene acuminolide (2) were isolated from the methanol extract of Vitex cofassus leaves. The chemical structure and the absolute configuration of 1 were determined by MS, NMR and electron circular dichroism (ECD) experiments. The isolated compounds were evaluated for their antiproliferative activities against a panel of human tumor cell lines, including a multidrug-resistant (MDR) cell line. Both compounds showed potent antiproliferative activities against all the tested cell lines with IC50 values of 5.4-11.4 µM. Their effec...

SC-CO2 extraction of Vitex agnus-castus L. fruits: The influence of pressure, temperature and water presoaking on the yield and GC –MS profiles of the extracts in comparison to the essential oil composition
The objectives were trifold: 1) to investigate the impact of pressure, temperature and water presoaking on SC-CO2 extraction of Vitex agnus-castus L. fruits with respect to the yield and GC–MS profiles of the extracts; 2) to find optimal extraction conditions by response surface methodology (RSM); 3) to compare GC–MS profiles of the extracts and essential oil. SC-CO2 extraction was performed at different pressures (8.4–36.6MPa) and temperatures (35.8–64.1°C) determined by central composite rotatable design. The essential oil was obtained by hydrodistillation. The extracts and oil were analyzed by GC–MS. Applied ...

Antibacterial activity of Vitex parviflora A. Juss. and Cyanthillium cinereum (L.) H. Rob. against human pathogens
This study investigated the antibacterial activities of methanolic crude extracts of Vitex parviflora (V. parviflora) leaves and stem and Cyanthillium cinereum (C. cinereum) leaves and roots. Crude methanolic extracts of V. parviflora and C. cinereum were tested for their antibacterial activities against Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa and Bacillus subtilis. The leaf and stem extracts of V. parviflora and root extracts of C. cinereum exhibited high antibacterial activity against Staphylococcus aureus. Increasing concentration of the V. parviflora leaves and roots crude extracts resulted in a...

Cardioprotective effect of Vitex negundo on isoproterenol-induced myocardial necrosis in wistar rats: A dual approach study.
Abstract Cardiovascular diseases (CVDs) are becoming the major cause of deaths worldwide, and their treatment demands novel therapeutic strategies. In this connection, we have identified p21 activated kinase 1 (PAK1) as a novel therapeutic target for the treatment of myocardial infarction (MI), where its expression is decreased when MI is induced with isoproterenol (ISO), which was brought back normal with pretreatment of Vitex negundo leaf ethanolic extract (VNE). These results were also supported by histopathological studies, cardiac markers, antioxidants, and inflammatory cytokines (NF-κB and IL-1β). ...
