GSE223796 Donor regulatory T cells rapidly adapt to recipient tissues to control acute graft-versus-host disease [bulk RNA-seq]
Contributors : David J Dittmar ; Michael RehliSeries Type : Expression profiling by high throughput sequencingOrganism : Mus musculusAdoptive transfer of donor regulatory T cells (Treg) is a promising treatment option for Graft-versus-Host disease (GvHD), but has not yet found its way into routine clinical practice. To map distinctive properties of protective Treg (generated by either polyclonal or allogeneic in vitro expansion), we followed their fate in recipient organs (spleen, liver, colon) in a prophylactic mouse model of MHC-mismatched bone-marrow transplantation (BMT). Using comprehensive gene expression and T cell...

RAGE in circulating immune cells is fundamental for hippocampal inflammation and cognitive decline in a mouse model of latent chronic inflammation
CONCLUSIONS: These findings indicate that RAGE in inflammatory cells is necessary to mediate stimuli of latent chronic inflammation that cause brain inflammation and cognitive decline, potentially by orchestrating monocyte activation via regulation of PSGL-1 expression. Our results also suggest esRAGE-mediated inflammatory regulation as a potential therapeutic option for cognitive dysfunction in MetS with latent chronic inflammation.PMID:38142917 | DOI:10.1016/j.bbi.2023.12.022 (Source: Brain, Behavior, and Immunity)

Inherited Human BCL10 Deficiencies
AbstractHuman BCL10 deficiency causes combined immunodeficiency with bone marrow transplantation as its only curative option. To date, there are four homozygous mutations described in the literature that were identified in four unrelated patients. Here, we describe a fifth patient with a novel mutation and summarize what we have learned about BCL10 deficiency. Due to the severity of the disease, accurate knowledge of its clinical and immunological characteristics is instrumental for early diagnosis and adequate clinical management of the patients. Clinical characteristics, disease trajectories and management of vacuoles, E1 enzyme, X-linked, autoinflammatory, somatic (VEXAS) syndrome: a systematic review
ConclusionVEXAS syndrome is a rare disorder affecting predominantly middle-aged men. This is the first systematic review to capture clinical manifestations, genetics and treatment of reported cases. Further studies are needed to optimise treatment and subsequently reduce morbidity and mortality. (Source: Rheumatology International)

Fighting viruses with viruses? ‘Gene drive’ offers new strategy to beat infections
If your immune system or drugs can’t stop a viral infection, why not pit a virus against itself? That’s the provocative idea several labs are pursuing. They are studying whether deliberately introducing engineered viruses into people infected with their natural relatives can “drive” a foreign gene into those viruses that ultimately wipes out an infection. No lab has knocked down an infection in animals this way yet, but a group has now shown it’s theoretically possible. These so-called gene drives harness the genome editor CRISPR to perform genetic surgery that speeds the spread of a gene through progeny. S...
