171TiPPhase II study of olaparib in previously treated advanced solid tumours with homologous recombination repair mutation (HRRm) or homologous recombination repair deficiency (HRD): LYNK-002

AbstractBackgroundPoly(ADP-ribose) polymerases (PARPs) play a key role in DNA damage repair (DDR). DDR defects due to HRRm (eg BRCA mutation [BRCAm]) or resulting in HRD (eg global loss of heterozygosity) may sensitize tumors to PARP inhibitors, eg olaparib. Olaparib monotherapy has activity in BRCAm ovarian, breast, pancreatic, and prostate cancer, and in prostate cancer with DDR defects beyond BRCAm (TOPARP). Olaparib improved efficacy outcomes vs placebo/chemotherapy as treatment in  ≤ 3rd-line (3L) HER2-negative BRCAm breast cancer (OlympiAD) and ≥2L BRCAm ovarian cancer (SOLO3), and maintenance therapy in ≥ 1L ovarian cancer irrespective of BRCAm (SOLO1, Study 19). Is there any difference in sensitivity to kidney function between men and women? Paterson et  al. have focused on sex differences in chronic kidney disease. Surprisingly, their experimental results show that only one microRNA, miR-146b-5p, affected the susceptibility of renocardiac pathology. They generated miR-146b knockout rats and found that miR-146b−/− females developed exacerbated renal hypertrophy and fibrosis and had less cardiac remodeling. Although miR-146b-5p has been reported to be upregulated in various types of cancers, this article reveals the novel role of miR-146b in the kidney.

The objective of this study is to assess information, motivation, and behavioral skills associated with willingness to receive an HPV test instead of a Pap test among women. The sample included U.S. 812 women, ages 30 to 65 years. Participants completed an online survey in 2018. ConclusionsOur results revealed specific details regarding residents’ error management strategies and provides validity evidence for the use of human factors error frameworks in surgical performance assessments. Practice in simulation-based learning environments may improve resident decision-making and error management opportunities by providing a structured experience where errors are explicitly characterized and used for training and feedback. Error management training may play a major role in equipping residents and junior faculty with the skills required for independent, high-quality operative performance.
