Characterisation of CYP2B6 and CYP2A6 pharmacogenetic variation in sub-Saharan African populations
This study therefore aimed to characterise the star allele (haplotype) distribution in CYP2B6 and CYP2A6 across diverse and understudied sub-Saharan African (SSA) populations. We called star alleles from 961 high-depth full genomes using StellarPGx, Aldy and PyPGx. In addition, we performed CYP2B6 and CYP2A6 star allele frequency comparisons between SSA and other global biogeographical groups represented in the new 1000 Genomes Project high-coverage dataset (n=2000). This study presents frequency information for star alleles in CYP2B6 (e.g. *6 and *18; frequency of 21-47% and 2-19%, respectively) and CYP2A6 (e.g. *4, *9 an...

Simultaneous quantification of 55 psychotropic drugs and metabolites in human plasma with a fast UPLC-MS/MS method
CONCLUSION: We developed and validated one of the most sensitive and complete UPLC-MS/MS methods in psychopharmacology, allowing the simultaneous determination of 55 psychotropic drugs in only 5.7 min after a simple sample preparation. This method has been successfully used in daily practice for therapeutic drug monitoring of psychiatric patients and is especially useful in polymedicated patients.PMID:38007935 | DOI:10.1016/j.biopha.2023.115918 (Source: Biomedicine and pharmacotherapy = Biomedecine and pharmacotherapie)

Ciprofol is primarily glucuronidated by UGT1A9 and predicted not to cause drug-drug interactions with typical substrates of CYP1A2, CYP2B6, and CYP2C19
This study systematically characterized UGTs that contribute to the formation of M4 using human liver microsomes (HLM), human intestinal microsomes (HIM), and human recombinant UGTs. The inhibitory potential of ciprofol and M4 against major human UGTs and cytochrome P450 enzymes (P450s) was also explored. In vitro-in vivo extrapolation (IVIVE) and physiologically-based pharmacokinetic (PBPK) simulations were performed to predict potential in vivo drug-drug interactions (DDIs) caused by ciprofol. Glucuronidation of ciprofol followed Michaelis-Menten kinetics in both HLM and HIM with apparent Km values of 345 and 412μM, Vma...

Real-world assessment of treatment inertia in the management of patients treated for major depressive disorder in the USA
Conclusion: Effective symptom management is critical for optimal outcomes in MDD. Results demonstrate that treatment inertia is common in MDD despite guidelines recommending treatment modification in patients not reaching remission.PMID:37987716 | DOI:10.57264/cer-2023-0091 (Source: Journal of Comparative Effectiveness Research)

Patient interest in acupuncture for smoking cessation: a survey
CONCLUSION: When comparing treatment options, patients reported more interest in acupuncture than other treatment options with a statistically significant difference in the level of interest between acupuncture and bupropion. All barriers (cost, time and effectiveness) were equally rated on a Likert-type scale with a median of 50 on a 101-point scale.PMID:37909713 | DOI:10.1177/09645284231206145 (Source: Acupuncture in Medicine : journal of the British Medical Acupuncture Society)

The effectiveness of the Intensive Tobacco Cessation Programme: a multi-professional approach
Conclusions: The number of cigarettes and nicotine dependence influence smoking cessation but individualised pharmacological treatment, an intensive behaviour change approach and multi-professional teamwork are the key to the success of an effective smoking cessation programme. Smoking cessation integrated with a lung cancer screening program: most effective method?
Conclusions: Smoking cessation and LCS associated seem to increase the probability of smoking cessation. It is recommended to associate a LCS program with a smoking cessation program as this encourages smoking cessation. (Source: European Respiratory Journal)

Effect of next-step antidepressant treatment on suicidal ideation: findings from the VAST-D trial
CONCLUSION: SI declines for most patients during next-step medication treatments. But about 1 in 5 experienced emergent or worsening SI during treatment, so vigilance for suicide risk through the entire 12-week acute treatment period is necessary. Treatment selection may affect the risk of SI.PMID:37859623 | DOI:10.1017/S0033291723003008 (Source: Psychological Medicine)

Increased depression-like behaviors with altered brain dopamine metabolisms in male mice housed in large cages are alleviated by bupropion
Eur J Pharmacol. 2023 Oct 17:176126. doi: 10.1016/j.ejphar.2023.176126. Online ahead of print.ABSTRACTPsycho-environmental stress-based animal models of anxiety and depression are useful for investigating pathological mechanisms and drug development. Although several rodent-based studies have reported the beneficial effects of environmental enrichment (EE) on brain plasticity and anxiety- and depression-like behaviors, other studies have reported inverse effects. Here, we found that housing male mice in EE involving large cages and other EE materials increased anxiety- and depression-like behaviors in open field and tail s...
