The primary role of sebum in the pathophysiology of acne vulgaris and its therapeutic relevance in acne management
CONCLUSIONS: To date, only combined oral contraceptives and oral spironolactone (both limited to female patients), oral isotretinoin and topical clascoterone (cortexolone 17α-propionate) modulate sebum production in patients with AV. A better understanding of mechanisms underlying sebaceous gland changes driving AV development is needed to expand the AV treatment armamentarium.PMID:38146664 | DOI:10.1080/09546634.2023.2296855 (Source: Journal of Dermatological Treatment)

Gene expression analysis to identify mechanisms underlying improvement of myocardial fibrosis by finerenone in SHR
Biochem Pharmacol. 2023 Dec 10:115975. doi: 10.1016/j.bcp.2023.115975. Online ahead of print.ABSTRACTBoth spironolactone and finerenone treatments significantly reduced SBP and there was no statistical difference in their antihypertensive effects. The differences in body weight (at the end of 1/2/3/4 week) to pre-dose body weight ratio and heart rate (at the end of 1/2/3/4 week) to pre-dose heart rate ratio were not statistically significant in the vehicle, spironolactone, finerenone, and control groups. There was no statistically significant difference in mortality among the vehicle, spironolactone, and finerenone groups. ...

Eco-friendly solvent bar microextraction based on a natural deep eutectic solvent and multivariate optimization for simultaneous determination of spironolactone and canrenone in urine and plasma samples
Environ Sci Pollut Res Int. 2023 Dec 11. doi: 10.1007/s11356-023-31217-w. Online ahead of print.ABSTRACTSimultaneous measurement of spironolactone and canrenone in urine and plasma provides valuable insight into renal function, and therapeutic efficacy and can be utilized to identify potential health risks and ensure patient safety throughout treatment. By adopting greener methods to analyze these compounds, significant reductions in the environmental impact of such studies can be achieved. For this purpose, a sensitive and eco-friendly solvent bar microextraction method using natural deep eutectic solvent (NDE) followed b...

Cost-effectiveness of Spironolactone for Adult Female Acne (SAFA): economic evaluation alongside a randomised controlled trial
Conclusions The results demonstrate a high level of uncertainty, particularly with respect to estimates of incremental QALYs. Compared with no active systemic treatment, spironolactone was estimated to be marginally cost-effective where multiple imputation was performed but was not cost-effective in complete case analysis. Pharmacokinetics of oral spironolactone in infants up to 2  years of age
ConclusionThe pharmacokinetics of spironolactone and its metabolites was highly variable between patients below 2  years of age. Body weight explained a significant part of this variability; this highlights the need to take it into account for dosing prescription in this population. Potentially inappropriate medicine use and predicting risk factors in hospitalized older adult patients: findings of a prospective observational study from Ethiopia
ConclusionsNearly three-fourths of the participants had potentially inappropriate medicine in their medicine list. Therefore, generating local evidence on the clinical, economic, and humanistic consequences may help in determining whether the Beers criteria should be taken into account when prescribing medicine to older adults. Interventions targeting older adult patients with heart failure might reduce inappropriate medicine use. (Source: Journal of Pharmaceutical Policy and Practice)

Increasing rates of referrals for investigation of primary aldosteronism at a tertiary centre
CONCLUSIONS: The rate of referrals and confirmatory testing for PA are increasing in our region. Adrenalectomy and mineralocorticoid antagonist therapy are both safe and effective treatments, although minor adverse effects were common with spironolactone.PMID:37956358 (Source: New Zealand Medical Journal)
