Nebulization of siRNA for inhalation therapy based on a microfluidic surface acoustic wave platform
Ultrason Sonochem. 2022 Jun 30;88:106088. doi: 10.1016/j.ultsonch.2022.106088. Online ahead of print.ABSTRACTThe local delivery of therapeutic small interfering RNA or siRNA to the lungs has the potential to improve the prognosis for patients suffering debilitating lung diseases. Recent advances in materials science have been aimed at addressing delivery challenges including biodistribution, bioavailability and cell internalization, but an equally important challenge to overcome is the development of an inhalation device that can deliver the siRNA effectively to the lung, without degrading the therapeutic itself. Here, we ...

Molecules, Vol. 27, Pages 4350: Melatonin Reverses the Warburg-Type Metabolism and Reduces Mitochondrial Membrane Potential of Ovarian Cancer Cells Independent of MT1 Receptor Activation
ustavo de Almeida Chuffa Ovarian cancer (OC) is the most lethal gynecologic malignancy, and melatonin has shown various antitumor properties. Herein, we investigated the influence of melatonin therapy on energy metabolism and mitochondrial integrity in SKOV-3 cells and tested whether its effects depended on MT1 receptor activation. SKOV-3 cells were exposed to different melatonin concentrations, and experimental groups were divided as to the presence of MT1 receptors (melatonin groups) or receptor absence by RNAi silencing (siRNA MT1+melatonin). Intracellular melatonin levels increased after treatment with melatonin in...

The role of the LINC01234/miR-433-3p/GRB2 ceRNA network in NSCLC cell malignant proliferation
CONCLUSION: LINC01234 competitively bound to miR-433-3p and promoted GRB2 transcription to augment NSCLC cell malignant proliferation.PMID:35747966 | DOI:10.2174/1386207325666220624093957

Emerging approaches for enabling RNAi therapeutics
Chem Asian J. 2022 Jun 11. doi: 10.1002/asia.202200451. Online ahead of print.ABSTRACTRNA interference (RNAi) is a primitive evolutionary mechanism developed to escape incorporation of foreign genetic material. siRNA has been instrumental in achieving the therapeutic potential of RNAi by theoretically silencing any gene of interest in a reversible and sequence-specific manner. Extrinsically administered siRNA generally needs a delivery vehicle to span across different physiological barriers and load into the RISC complex in the cytoplasm in its functional form to show its efficacy. This review discusses the designing princ...

Molecules, Vol. 27, Pages 3151: Neuroprotective Effect of Dioscin against Parkinson & rsquo;s Disease via Adjusting Dual-Specificity phosphatase 6(DUSP6)-Mediated Oxidative Stress
In this study, the tests on 6-hydroxydopamine (6-OHDA)-induced PC12 cells and rats were carried out. The results showed that dioscin dramatically improved cell viability, decreased reactive oxygen species (ROS) levels, improved motor behavior and tyrosine hydroxylase(TH) levels and restored the levels of glutathione (GSH) and malondialdehyde (MDA) in rats. Mechanism investigation showed that dioscin not only markedly increased the expression level of dual- specificity phosphatase 6 (DUSP6) by 1.87-fold in cells and 2.56-fold in rats, and decreased phospho-extracellular regulated protein kinases (p-ERK) level by 2.12-fold i...

Effects of long noncoding RNA AK093407 on the biological behavior of colon cancer cells and the underlying mechanism
CONCLUSION: These results showed that knockdown of AK093407 could inhibit colon cancer cell proliferation, induce apoptosis and cell cycle arrest, influence the expression of vital factors in mitochondrial apoptosis pathway and cell cycle regulatory pathway, and may negatively regulate JAK/STAT3 through down-regulating p-Stat3.PMID:35400339 | DOI:10.2174/1386207325666220408092028
