TXNDC5 Contributes to Rheumatoid Arthritis by Down-regulating IGFBP1 Expression.
This study investigated whether TXNDC5 contributes to RA via the insulin signaling pathway. In the study, RA synovial fibroblast-like cells (RASFs) transfected with an anti-TXNDC5 small interfering RNA (siRNA) were analyzed with an Insulin Signaling Pathway RT(2) Profiler PCR Array and an Insulin Resistance RT(2) Profiler PCR Array. The PCR arrays detected significantly increased expression of insulin-like growth factor binding protein 1 (IGFBP1) in RASFs with suppressed TXNDC5 expression. The result was verified using real-time PCR and western blot analyses. Significantly elevated IGFBP1 expression and decreased IL-6 secr...

Targeted Combination of Antioxidative and Anti ‐Inflammatory Therapy of Rheumatoid Arthritis using Multifunctional Dendrimer‐Entrapped Gold Nanoparticles as a Platform
A multifunctional dendrimer nanoplatform is developed to have entrapped gold nanoparticles within dendrimer interiors, and covalently linked PEGylated alpha ‐tocopheryl succinate, PEGylated folic acid, and 1,3‐propane sultone on dendrimer peripheries. The designed nanoplatform enables co‐delivery of drug/gene to macrophage cells, effectively scavenging reactive oxygen species and inhibiting tumor necrosis factor‐α expression for combined antiox idative and anti‐inflammatory therapy of rheumatoid arthritis. AbstractAbundant reactive oxygen species and tumor necrosis factor ‐α (TNF‐α) cytokine supply of M1...

Notch ‐1 and Notch‐3 Mediate Hypoxia‐Induced Activation of Synovial Fibroblasts in Rheumatoid Arthritis
ConclusionThis study identified a functional link between HIF-1 α, Notch-1, and Notch-3 signaling in regulating activation of RASFs and the processes involved in the pathogenesis of RA.

KDM6B epigenetically regulated-interleukin-6 expression in the dorsal root ganglia and spinal dorsal horn contributes to the development and maintenance of neuropathic pain following peripheral nerve injury in male rats
Brain Behav Immun. 2021 Aug 28;98:265-282. doi: 10.1016/j.bbi.2021.08.231. Online ahead of print.ABSTRACTThe lysine specific demethylase 6B (KDM6B) has been implicated as a coregulator in the expression of proinflammatory mediators, and in the pathogenesis of inflammatory and arthritic pain. However, the role of KDM6B in neuropathic pain has yet to be studied. In the current study, the neuropathic pain was determined by assessing the paw withdrawal threshold (PWT) and paw withdrawal latency (PWL) following lumbar 5 spinal nerve ligation (SNL) in male rats. Immunohistochemistry, Western blotting, qRT-PCR, and chromatin immu...

Extracellular sulfatase-2 is overexpressed in rheumatoid arthritis and mediates the TNF- α-induced inflammatory activation of synovial fibroblasts
This study identified a novel role of Sulf-2 in TNF-α signaling and the activation of RA synoviocytes, providing the rationale for evaluating the therapeutic targeting of Sulf-2 in preclinical models of RA.PMID:36068294 | DOI:10.1038/s41423-022-00913-x

Interleukin-13 reduces bone erosion in rheumatoid arthritis by up-regulating osteoprotegerin expression in fibroblast-like synoviocytes : an in vitro and in vivo study
CONCLUSIONS: IL-13 can inhibit osteoclastogenesis by up-regulating OPG in RA-FLSs through IL-13 receptors via the STAT6 pathway, thus may ameliorate bone erosion in RA.PMID:36995338 | DOI:10.55563/clinexprheumatol/b96n1e
