Nek2 siRNA therapy using a portal venous port –catheter system for liver metastasis in pancreatic cancer
In this study, the overexpression of Nek2 was identified in pancreatic cancer cell lines. Nek2 siRNA inhibited tumor growth in a subcutaneous xenograft mouse model of pancreatic cancer, prolonged the survival time in an intraperitoneal xenograft mouse model and efficiently prevented the progression of liver metastasis using a portal venous port–catheter system. Taken together, Nek2 is an effective therapeutic target in pancreatic cancer. An adequate delivery system is considered important in treating advanced pancreatic cancer, such as peritoneal dissemination and liver metastasis. Further investigations are required on ...

cyp51A gene silencing using RNA interference in azole‐resistant Aspergillus fumigatus
Summary An increasing number of reports have described the emergence of acquired resistance of Aspergillus fumigatus to azole compounds. The primary mechanism of resistance in clinical isolates is the mutation of the azole drug target enzyme, which is encoded by the cyp51A gene. The aim of this study was to evaluate the impact of silencing the cyp51A gene in azole‐resistant A. fumigatus isolates. A 21‐nucleotide small‐interfering RNA (siRNA) was designed based on the cDNA sequence of the A. fumigatus cyp51A gene. After silencing the cyp51A gene in germinated conidia (15, 20, 25 and 50 nM), azole‐resistant A. fumiga...

HMGB1 mediates Aspergillus fumigatus-induced inflammatory response in alveolar macrophages of COPD mice via activating MyD88/NF- κB and syk/PI3K signalings.
In conclusion, HMGB1 is responsible for A. fumigatus-induced inflammatory response in COPD alveolar macrophage via Dectin-1 and TLR2/4 receptor through activating MyD88/NF-κB and syk/PI3K signalings. PMID: 29078091 [PubMed - as supplied by publisher]

Galectin-3 plays an important pro-inflammatory role in A. fumigatus keratitis by recruiting neutrophils and activating p38 in neutrophils
CONCLUSION: These data proved that A. fumigatus increased Gal-3 expression and elevated disease clinical scores, PMNs infiltration and cytokines expression through Gal-3. In PMNs, A. fumigatus upregulated IL-1β and IL-6 secretion through the Gal-3 / p38 pathway.PMID:33933850 | DOI:10.1016/j.intimp.2021.107706

Functional role of protein kinase D1 in Aspergillums fumigates-induced activation of nuclear factor-κB signal pathway and transcription.
CONCLUSION: PKD1 may contribute to the activation of NF-κB signal pathway and NF-κB transcription induced by Aspergillums fumigates. PMID: 23529230 [PubMed - in process]

Alternaria Induces Production of Thymic Stromal Lymphopoietin in Nasal Fibroblasts Through Toll-like Receptor 2.
CONCLUSIONS: The results of this study show that TSLP expression could be induced in nasal fibroblasts by exposure to Alternaria and that TLR2 may be involved in the process. The promotion of TSLP production in nasal fibroblasts by airborne fungi may facilitate the development or exacerbation of Th2-type nasal inflammation, especially in CRS with nasal polyps. Aspergillus fumigatus promotes T helper type 2 responses through thymic stromal lymphopoietin production by human corneal epithelial cells
ConclusionThese findings suggested that HCEC‐derived TSLP has a key role in adaptive immune responses of fungal keratitis via skewing Th2 differentiation and promoting humoral immunity. This article is protected by copyright. All rights reserved.

Dectin-1 is essential for IL-1 β production through JNK activation and apoptosis in Aspergillus fumigatus keratitis.
CONCLUSIONS: Being an important inflammatory PRR to mediate host inflammatory response, Dectin-1 induced IL-1β production is JNK dependent in mouse A. fumigatus keratitis, and suppressed apoptosis mediated anti-inflammatory response. PMID: 28926773 [PubMed - as supplied by publisher]

GSDMD, an executor of pyroptosis, is involved in IL-1 β secretion in Aspergillus fumigatus keratitis.
GSDMD, an executor of pyroptosis, is involved in IL-1β secretion in Aspergillus fumigatus keratitis. Exp Eye Res. 2020 Dec 03;:108375 Authors: Zhao W, Yang H, Lyu L, Zhang J, Xu Q, Jiang N, Liu G, Wang L, Yan H, Che C Abstract The protein GSDMD is an important performer of pyroptosis and a universal substrate for the inflammatory caspase. However, the role and regulatory mechanism of GSDMD in Aspergillus fumigatus keratitis is remains unknown. Here we detected GSDMD protein in the cornea of normal and fungal-infected C57BL/6 mice. Human corneal epithelial cell (HCECs) were preincubated with a hydroch...
