MiR146a modulates chondrogenesis of bone marrow mesenchymal stem cells by modulating Lsm11 expression
This study aimed to determine the function of miR146a in the chondrogenic differentiation of MSCs and the potential mechanisms involved. MiR146a expression increased during chondrogenesis. MiR146a knockout (KO) led to the increased chondrogenesis of MSCs compared to that in wild-type (WT) MSCs, whereas the overexpression of miR146a by mimics resulted in the decreased chondrogenesis of MSCs, as determined by the mRNA expression of collagen, type II, alpha 1 (COL2A1), aggrecan, cartilage oligomeric matrix protein (COMP), and matrix metallopeptidase 13 (MMP13). Furthermore, cartilage defects could be treated better when injec...

A20 binding and inhibitor of nuclear factor kappa B (NF- κB)-1 (ABIN-1) - a novel modulator of mitochondrial autophagy
Am J Physiol Cell Physiol. 2022 Nov 28. doi: 10.1152/ajpcell.00493.2022. Online ahead of print.ABSTRACTA20 binding inhibitor of nuclear factor kappa B (NF-κB)-1 (ABIN-1), a polyubiquitin-binding protein, is a signal-induced autophagy receptor that attenuates NF-κB-mediated inflammation and cell death. The present study aimed to elucidate the potential role of ABIN-1 in mitophagy, a biological process whose outcome is decisive in diverse physiological and pathological settings. Microtubule-associated proteins 1A/1B light chain 3B-II (LC3B-II) was found to be in complex with ectopically expressed hemagglutinin (HA)-tagged-...

FXYD3 facilitates Na < sup > + < /sup > and liquid absorption across human airway epithelia by increasing the transport capacity of the Na/K ATPase
Am J Physiol Cell Physiol. 2022 Aug 22. doi: 10.1152/ajpcell.00047.2022. Online ahead of print.ABSTRACTNa/K ATPase activity is essential for ion transport across epithelia. FXYD3, a ᵯE; subunit of the Na/K ATPase, is expressed in the airway, but its function remains undetermined. Single cell RNA sequencing and immunohistochemistry revealed that FXYD3 localizes within the basolateral membrane of all airway epithelial cells. To study FXYD3 function, we reduced FXYD3 expression using siRNA. After permeabilizing the apical membrane with nystatin, epithelia pretreated with FXYD3-targeting siRNA had lower ouabain-sensitive sho...

Mitofusin 2 positively regulates Ca < sup > 2+ < /sup > signaling by tethering the sarcoplasmic reticulum and mitochondria in rat aortic smooth muscle cells
This study strongly suggests that Mfn2 is responsible for SR-mitochondria Ca2+ signaling by tethering mitochondria to SR, thereby regulating ATP production and proliferation of VSMCs.PMID:35704692 | DOI:10.1152/ajpcell.00274.2021
