MARCH8 downregulation modulates profibrotic responses including myofibroblast differentiation
In this study, we demonstrated decreased MARCH8 expression in IPF patients compared to non-PF controls and in bleomycin induced PF. TGF-β dose and time dependently decreased MARCH8 expression in normal and IPF human lung fibroblast (HLFs), along with induction of FMT markers α-SMA, collagen type I (Col-1), and fibronectin (FN). Interestingly, overexpression of MARCH8 significantly suppressed TGF-β-induced expression of α-SMA, Col-1, and FN. By contrast, knockdown of MARCH8 using siRNA upregulated basal expression of α-SMA/Col-1/FN. Moreover, MARCH8 knockdown enhanced TGF-β-induced FMT marker expression. These data cl...

Long non-coding RNA TUG1 regulates smooth muscle cell differentiation via KLF4-Myocardin axis
Am J Physiol Cell Physiol. 2023 Aug 29. doi: 10.1152/ajpcell.00275.2023. Online ahead of print.ABSTRACTAbdominal Aortic Aneurysms (AAAs) are asymptomatic vascular diseases that have life-threatening outcomes. Smooth-muscle cell (SMC) dysfunction plays an important role in AAA development. The contribution of non-coding genome, specifically the role of long non-coding RNAs (lncRNAs) in SMC dysfunction is relatively unexplored. we investigated the role of lncRNA TUG1 in SMC dysfunction. To identify potential lncRNAs relevant to SMC functionality, lncRNA profiling was performed in angiotensin-II treated SMCs. AAA was induced ...

MicroRNA-140 is not involved in sepsis-induced muscle atrophy
Am J Physiol Cell Physiol. 2023 Jul 24. doi: 10.1152/ajpcell.00157.2023. Online ahead of print.ABSTRACTSepsis is a life-threatening inflammatory response to infection, often accompanied by skeletal muscle atrophy. A previous study demonstrated that the administration of microRNA-140 (miR-140) attenuated lipopolysaccharides (LPS)-induced muscle atrophy, whereas miR-140 knockdown with siRNA promoted atrophy. Therefore, we investigated whether miR-140 is involved in LPS-induced muscle atrophy using a genetic model, miR-140-/- mice. We found that a single injection of LPS induced atrophy both in slow-twitch and fast-twitch mus...
