LncRNA MRAK159688 facilitates morphine tolerance by promoting REST-mediated inhibition of mu opioid receptor in rats
Neuropharmacology. 2022 Jan 1;206:108938. doi: 10.1016/j.neuropharm.2021.108938. Online ahead of print.ABSTRACTMorphine tolerance (MT) caused by the long-term use of morphine is a major medical problem. The molecular mechanism of morphine tolerance remains elusive. Here, we established a morphine tolerance model in rats and verified whether the long noncoding RNA (lncRNA) MRAK159688 is involved in morphine tolerance and its specific molecular mechanism. We show the significant upregulation of MRAK159688 expression in the spinal cord of morphine-tolerant rats. Overexpression of MRAK159688 by a lentivirus reduces the analges...

KDM6B epigenetically regulated-interleukin-6 expression in the dorsal root ganglia and spinal dorsal horn contributes to the development and maintenance of neuropathic pain following peripheral nerve injury in male rats
Brain Behav Immun. 2021 Aug 28;98:265-282. doi: 10.1016/j.bbi.2021.08.231. Online ahead of print.ABSTRACTThe lysine specific demethylase 6B (KDM6B) has been implicated as a coregulator in the expression of proinflammatory mediators, and in the pathogenesis of inflammatory and arthritic pain. However, the role of KDM6B in neuropathic pain has yet to be studied. In the current study, the neuropathic pain was determined by assessing the paw withdrawal threshold (PWT) and paw withdrawal latency (PWL) following lumbar 5 spinal nerve ligation (SNL) in male rats. Immunohistochemistry, Western blotting, qRT-PCR, and chromatin immu...

GSE174741 LINC01133 inhibits invasion and promotes proliferation in an endometriosis epithelial cell line
Contributors : Quanah J Hudson ; Iveta Yotova ; Florian M Pauler ; Rene WenzlSeries Type : Expression profiling by high throughput sequencingOrganism : Homo sapiensEndometriosis is a common gynecological disorder characterized by ectopic growth of endometrium outside the uterus and associated with chronic pain and infertility. We investigated the role of LINC01133 in endometriosis, an lncRNA that has been implicated in several types of cancer. We found that LINC01133 is upregulated in ectopic endometriosis lesions. As expression appeared higher in the epithelial endometrial layer, we performed an siRNA knockdown of LINC011...

Identification of lncRNA and mRNA expression profiles in dorsal root ganglion in rats with cancer-induced bone pain
CONCLUSION: LncRNA played important roles in regulation of CIBP or mRNA expression in peripheral neuropathy in CIBP. These alterd mRNAs and lncRNAs might be potential therapeutic targets for the treatment of CIBP.PMID:34364296 | DOI:10.1016/j.bbrc.2021.07.063

Secreted PLA < sub > 2 < /sub > -III is a possible therapeutic target to treat neuropathic pain
Biochem Biophys Res Commun. 2021 Jul 5;568:167-173. doi: 10.1016/j.bbrc.2021.06.058. Online ahead of print.ABSTRACTLysophosphatidic acid (LPA) plays a critical role in developing and maintaining chronic pain in various animal models. Previous studies have reported that cytosolic and calcium-independent phospholipase A2 (PLA2) is involved in the LPA receptor-mediated amplification of LPA production in the spinal dorsal horn (SDH) after nerve injury, while the involvement of secreted PLA2 (sPLA2) remains unclear. The present study revealed that only sPLA2 -III among 11 species of PLA2 showed a significant upregulation of gen...

ZEB1 Induces Ddr1  Promoter Hypermethylation and Contributes to the Chronic Pain in Spinal Cord in Rats Following Oxaliplatin Treatment
In conclusion, these results suggested that the ZEB1 recruited DNMT3b to the Ddr1 promoter, which induced the DDR1 downregulation and contributed to the oxaliplatin-induced chronic pain.PMID:34032956 | DOI:10.1007/s11064-021-03355-5

BDNF Participates in Chronic Constriction Injury-Induced Neuropathic Pain via Transcriptionally Activating P2X 7 in Primary Sensory Neurons
In this study, we demonstrated that the expression level of brain-derived neurotrophic factor (BDNF) was remarkably and time-dependently increased in dorsal root ganglion (DRG) neurons. DRG microinjection of BDNF siRNA in DRG ameliorated chronic constriction injury (CCI) induced mechanical, thermal, and cold nociceptive hypersensitivities. Overexpressing BDNF through microinjection of the AAV5-BDNF in DRG caused enhanced responses to basal mechanical, thermal, and cold stimuli in mice exposed to CCI. Mechanically, the P2X7 promoter activity was enhanced by CCI-induced increase of DRG BDNF protein and was involved in the CC...

TRAF6 Contributes to CFA-Induced Spinal Microglial Activation and Chronic Inflammatory Pain in Mice
In this study, we investigated the role and mechanism of TRAF6 in complete Freund's adjuvant (CFA)-evoked chronic inflammatory hypersensitivity and the effect of docosahexaenoic acid (DHA) on TRAF6 expression and inflammatory pain. We found that TRAF6 was dominantly increased in microglia at the spinal level after intraplantar injection of CFA. Intrathecal TRAF6 siRNA alleviated CFA-triggered allodynia and reversed the upregulation of IBA-1 (microglia marker). In addition, intrathecal administration of DHA inhibited CFA-induced upregulation of TRAF6 and IBA-1 in the spinal cord and attenuated CFA-evoked mechanical allodyni...

Duloxetine suppresses BMP-4-induced release of osteoprotegerin via inhibition of the SMAD signaling pathway in osteoblasts
Biochem Cell Biol. 2021 Mar 1. doi: 10.1139/bcb-2020-0347. Online ahead of print.ABSTRACTDuloxetine, a selective serotonin-norepinephrine reuptake inhibitor, is currently recommended to use in the treatment of chronic painful disorders such as fibromyalgia, chronic musculoskeletal pain and diabetic peripheral neuropathy. We previously demonstrated that bone morphogenetic protein-4 (BMP-4) stimulates osteoprotegerin (OPG) production in osteoblast-like MC3T3-E1 cells, and that p70 S6 kinase positively regulates the OPG synthesis. The present study is aimed to investigate the effect of duloxetine on BMP-4-stimulated OPG synth...
