Keloid Core Factor CTRP3 Overexpression Significantly Controlled TGF- < em > β < /em > 1-Induced Propagation and Migration in Keloid Fibroblasts
CONCLUSION: CTRP3 exerted an antifibrotic role through inhibiting proliferation, migration, and ECM accumulation of KFs via regulating TGF-β1/Smad signal path.PMID:37091895 | PMC:PMC10115533 | DOI:10.1155/2023/9638322

The molecular mechanism of GADD153 in apoptosis of keloid fibroblasts exposed to botulinum toxin type A
J Cell Mol Med. 2021 Sep 2. doi: 10.1111/jcmm.16881. Online ahead of print.ABSTRACTApoptosis plays a key role in keloids. Growth arrest and DNA damage-inducible gene 153 (GADD153) is regulated by apoptosis. Botulinum toxin type A (BTXA) can induce apoptosis in keloid fibroblasts. This research aimed to explore the hypothesis that GADD153 mediates apoptosis in keloid fibroblasts exposed to BTXA. BTXA significantly induced GADD153 protein and mRNA expression in keloid fibroblasts. Treatment with c-Jun N-terminal kinase (JNK) inhibitor SP600125, JNK small interfering RNA (siRNA) and tumour necrosis factor-alpha (TNF-α) antib...

Association between MeCP2 and Smad7 in the pathogenesis and development of pathological scars.
Authors: Li D, Yang E, Zhao J, Zhang H Abstract To explore the relationship between methylated binding protein 2 (MeCP2) and mothers against decapentaplegic homolog 7 (Smad7) in the pathogenesis and development of pathological scars. Immunohistochemistry, Western blot and real-time polymerase chain reaction (RT-PCR) were used to detect the expression of MeCP2 in different types of human scars and hypertrophic scars at different growth times. The methylation status of Smad7 gene promoter in different scar tissues was determined by methylation-specific PCR. After transfection with MeCP2-siRNA (small interfering RNA) ...

Treatment of keloids through Runx2 siRNA ‑induced inhibition of the PI3K/AKT signaling pathway.
In conclusion, the findings of the present study indicated that Runx2 silencing in HKFs might significantly inhibit the cell proliferation, migration and the expression levels of ECM‑related proteins, and promote apoptosis via suppressing the PI3K/AKT signaling pathway. Thus, Runx2 siRNA treatment may reverse the pathological phenotype of keloids through the inhibition of PI3K/AKT signaling in patients. PMID: 33200804 [PubMed - in process]

Resveratrol inhibits proliferation and promotes apoptosis of keloid fibroblasts by targeting HIF-1 α.
In this study, keloid-derived fibroblasts were cultured under hypoxia environment and was treated by resveratrol. CCK-8 assay and Annexin V-FITC were used to evaluate cell activity and apoptosis level. Western blot and RT-qPCR were also used to assess the expression of HIF-α, Collagen I and Collagen III. Besides, siRNA was also used to explore the mechanisms of resveratrol's effect. In this study, hypoxia promotes proliferation and inhibits apoptosis of keloid fibroblasts. These findings highlight the potential obstacle in treating keloids. Furthermore, we demonstrated that resveratrol could reverse the effect of hypoxia ...

Syndecan-1 regulates extracellular matrix expression in keloid fibroblasts via TGF-β1/Smad and MAPK signaling pathways
Publication date: Available online 15 January 2020Source: Life SciencesAuthor(s): Jing Cui, Shan Jin, Chenglong Jin, Zhehu JinAbstractAimsThe present study aimed to explore the effect of syndecan-1 on keloid fibroblasts.Main methodsImmunohistochemistry and Western blot were employed to assess the expression of syndecan-1. Primary cultured keloid fibroblasts were transfected with syndecan-1 siRNA. The function of syndecan-1 on the proliferation of keloid fibroblasts was investigated through Cell Counting Kit-8 (CCK-8) and flow cytometry. Extracellular matrix, TGF-β1/Smad, and MAPK related proteins were evaluated by Western...

Wnt2 knock down by RNAi inhibits the proliferation of in vitro-cultured human keloid fibroblasts
To study the effect of knocking down wingless-related MMTV integration site 2 (Wnt2) expression by RNAi on the growth and signaling pathways of ex vitro-cultured keloid fibroblasts (KFB). Human KFB were isolated from 10 keloid patient specimens. The KFB cells were then transfected with 4 pairs of small interfering RNA (siRNA) targeting human Wnt2, respectively. Reverse transcriptase-polymerase chain reaction and Western blot analysis were conducted to verify the knock down of Wnt2, and the expression of β-catenin glycogen synthase kinase-3β (GSK-3β) and cyclin D1 were examined. siRNA Wnt2 transfection (siWnt2) resulte...

1063 Topically applied NanoFlares to measure gene expression in vivo: Proof-of-concept
Spherical nucleic acids (SNAs) are topical agents that penetrate human skin to knock down genes via antisense and siRNA pathways. NanoFlares, SNAs that release sequence-specific fluorescence when binding to intracellular mRNA targets, measure mRNA levels in live cells. We hypothesized that NanoFlares could be topically applied to measure gene expression in vivo. As proof-of-concept, we detected connective tissue growth factor (CTGF), known to be increased in hypertrophic scars and keloids. Using fibroblasts from normal skin, keloids, hypertrophic scars and peri-scar tissue, we found that: a) NanoFlares are specific; b) exp...
