The heparan sulphate proteoglycan Syndecan ‐1 (CD138) regulates tumour progression in a 3D model of ductal carcinoma in situ of the breast
In this study, we employed a siRNA knockdown approach in the DCIS model cell line MCF10A DCIS.com to investigate a potential connection between Sdc-1 and epithelial mesenchymal transition (EMT), proteolysis and the Rho kinase pathway. Analysis of gene expression data of the TNMplot.com database revealed that Sdc-1 expression was higher in primary breast tumours compared to metastases. The impact of Sdc-1-depletion on the cellular phenotype was investigated in a Matrigel-based three-dimensional cell culture model. Sdc-1 depletion resulted in the formation of larger spheroids and the formation of invasive protrusions. Applic...

Role of the prorenin receptor in endometrial cancer cell growth
Oncotarget. 2022 Apr 1;13:587-599. doi: 10.18632/oncotarget.28224. eCollection 2022.ABSTRACTEndometrial cancer is the most diagnosed gynecological malignancy. Despite numerous scientific advances, the incidence and mortality rate of endometrial cancer continues to rise. Emerging evidence suggests a putative role of the (pro)renin receptor ((P)RR), in the ontogenesis of endometrial cancer. The (P)RR is implicated in breast cancer and pancreatic carcinoma pathophysiology by virtue of its role in proliferation, angiogenesis, fibrosis, migration and invasion. Thus, we aimed to investigate the functional role of the (P)RR in hu...

Syndecan-1 and -4 influence Wnt signaling and cell migration in human breast cancers
This study examined syndecan-1 (SDC1) and syndecan-4 (SDC4; SDC1/4) in breast cancer (BC) in vitro cell models and their role in tumorigenesis. Gene expression of HSPG core proteins, biosynthesis and modification enzymes along with Wnt/FGF morphogen pathway components were examined following inhibition of SDC1 and SDC4 via small interfering RNA (siRNA), and enhancement of HSPGs via addition of heparin and FGF. siRNAs knockdowns (KDs) were performed in the MCF-7 (lowly invasive and poorly metastatic) and the MDA-MB-231 (highly invasive and metastatic) human BC cell lines. Significantly decreased gene expression of SDC1 and ...

LncRNA KRT19P3 Is Involved in Breast Cancer Cell Proliferation, Migration and Invasion
In conclusion, this study demonstrated that lncRNA KRT19P3 inhibits BC progression, and may affect the expression of PD-L1 in BC, which in turn affects CD8+ T (CD8 positive Cytotoxic T lymphocyte) cells in the immune microenvironment.

Docetaxel-triggered SIDT2/NOX4/JNK/HuR signaling axis is associated with TNF- α-mediated apoptosis of cancer cells
Biochem Pharmacol. 2021 Dec 2:114865. doi: 10.1016/j.bcp.2021.114865. Online ahead of print.ABSTRACTPrevious studies have confirmed that docetaxel (DTX) treatment increases TNF-α production in cancer cells, but its mechanism of action remains unclear. Therefore, this study aimed to determine the signaling axis by which DTX induced the expression of TNF-α in U937 leukemia and MCF-7 breast carcinoma cells. DTX treatment promoted Ca2+-controlled autophagy and SIDT2 expression, resulting in lysosomal degradation of miR-25 in U937 cells. Downregulation of miR-25 increased NOX4 mRNA stability and protein expression. NOX4-stimu...
