Direct cytosolic siRNA delivery by reconstituted high density lipoprotein for target-specific therapy of tumor angiogenesis.
We described here the mechanisms by which small interfering RNA (siRNA) molecules incorporated in reconstituted high density lipoprotein (rHDL) were efficiently transferred into the cytoplasm of cells to perform target-specific therapy of tumor angiogenesis. Using fluorescent-tagged apolipoprotein A-I (apoA-I) and cholesterol-conjugated siRNA (Chol-siRNA), it was confirmed with FACS and confocal microscopic measurements that Chol-siRNA-loaded rHDL nanoparticles (rHDL/Chol-siRNA complexes) were successfully established and apoA-I certainly was attached to the surface of Chol-siRNA-loaded lipoplexes (Lipos/Chol-siRNA complex...

Functionalized selenium nanoparticles for targeted siRNA delivery silence Derlin1 and promote antitumor efficacy against cervical cancer.
In this study, biocompatible selenium nanoparticles (SeNPs) were loaded with RGDfC peptide to fabricate tumor-targeting gene delivery vehicle RGDfC-SeNPs. Subsequently, RGDfC-SeNPs were loaded with Derlin1-siRNA to fabricate RGDfC-Se@siRNA, which are functionalized selenium nanoparticles. RGDfC-Se@siRNA showed greater uptake in HeLa cervical cancer cells in comparison with that in human umbilical vein endothelial cells (HUVECs), verifying the RGDfC-mediated specific uptake of RGDfC-Se@siRNA. RGDfC-Se@siRNA was capable of entering HeLa cells via clathrin-associated endocytosis, and showed faster siRNA release in a cancer ce...

Unprotonatable and ROS ‐Sensitive Nanocarrier for NIR Spatially Activated siRNA Therapy with Synergistic Drug Effect
An unprotonatable and reactive oxygen species-responsive nanocarrier is designed to co-deliver photodynamic reagent chlorin e6 (Ce6) and small interfering RNA (siRNA). Owing to the unprotonatable property and near infrared-activated photochemical internalization effect, the cationic nanocarrier spatially controls siRNA activity “on/off”, turning on siRNA therapy with the synergistic effect of Ce6-based photodynamic therapy in tumor tissue and turning off siRNA activity in normal tissues. AbstractAlthough small interfering RNA (siRNA) therapy has achieved great progress, unwanted gene inhibition in normal tissues severe...

GSE87497 Evaluating Side Effects of Nanoparticle-mediated siRNA Delivery to Mesenchymal Stem Cells Using Next-generation Sequencing and Enrichment Analysis
This study investigates both intended and unintended effects of diblock copolymer nanoparticle (NP) delivery of siRNA delivery to human mesenchymal stem cells (hMSC). Specifically, siRNA delivery was investigated at a range of NP-siRNA:hMSC ratios with a focus on the effects of NP-siRNA treatment on hMSC functions. Additionally, next-generation RNA sequencing (RNA-seq) was used with enrichment analysis to observe side effects in hMSC gene expression. Results show NP-siRNA delivery is negatively correlated with hMSC density. However, higher NP-siRNA:hMSC ratios increased cytotoxicity and decreased metabolic activity. hMSC p...

Design of a Novel PEGylated Liposomal Vector for Systemic Delivery of siRNA to Solid Tumors.
Abstract A small interfering RNA (siRNA) delivery system using dioleylphosphate-diethylenetriamine conjugate (DOP-DETA)-based liposomes (DL) was assessed for systemic delivery of siRNA to tumors. DL carrying siRNA capable of inducing efficient gene silencing with low doses of siRNA were modified with polyethylene glycol (PEG-DL/siRNA) for systemic injection of siRNA. The biodistribution of DL and siRNA in the PEG-DL/siRNA was studied by using radiolabeled DL and fluorescence-labeled siRNA, respectively. DL in the PEG-DL/siRNA showed a high retention in the plasma, accumulation in the tumor, and low accumulation in...

Multiwalled carbon nanotubes co-delivering sorafenib and epidermal growth factor receptor siRNA enhanced tumor-suppressing effect on liver cancer.
CONCLUSIONS: MWNT/Sor/siRNA exhibited a superior antitumor effect in vitro and in vivo. METHODS: The MWNT/Sor and MWNT/Sor/siRNA were prepared, and then the morphologies of MWNT/Sor/siRNA were analyzed. In vitro Sor release assay, siRNA stability and cellular uptake of MWNT/Sor/siRNA were performed as well. Next, the effects of MWNT, free Sor, free siRNA, MWNT/Sor and MWNT/Sor/siRNA were evaluated by colony-forming assay, and cell apoptosis assay in HepG2 cells. Meanwhile, the level of EGFR and proteins associated with apoptosis was tested. Furthermore, the anti-tumor effects of MWNT/Sor/siRNA on LC xenograft mice wer...

INFLUENCE OF mRNA FEATURES ON siRNA INTERFERENCE EFFICACY.
Abstract Design of small interference RNA (siRNA) is one of the most important steps in effectively applying the RNA interference (RNAi) technology. The current siRNA design often produces inconsistent design results, which often fail to reliably select siRNA with clear silencing effects. We propose that when designing siRNA, one should consider mRNA global features and near siRNA-binding site local features. By a linear regression study, we discovered strong correlations between inhibitory efficacy and both mRNA global features and neighboring local features. This paper shows that, on average, less GC content, fe...

Enhanced silencing and stabilization of siRNA polyplexes by histidine-mediated hydrogen bonds.
Abstract Branched peptides containing histidines and lysines (HK) have been shown to be effective carriers for DNA and siRNA. We anticipate that elucidation of the binding mechanism of HK with siRNA will provide greater insight into the self-assembly and delivery of the HK:siRNA polyplex. Non-covalent bonds between histidine residues and nucleic acids may enhance the stability of siRNA polyplexes. We first compared the polyplex biophysical properties of a branched HK with those of branched asparagine-lysine peptide (NK). Consistent with siRNA silencing experiments, gel electrophoresis demonstrated that the HK siRN...

A New Approach of Delivering siRNA to the Cornea and its Application for Inhibiting Herpes Simplex Keratitis.
Abstract Small interfering RNA (siRNA) is a potential agent for the treatment of ocular surface diseases. Previous studies delivered siRNA by directly injecting siRNA into cornea or conjunctiva. In the present study we sought to explore an alternative approach to deliver siRNA into mouse cornea via eye drops that contains cy3-labeled siRNA (cy3-siRNA) and different cationic complexing agents and to evaluate the effects of siRNA targeting HSV-1 ICP4 gene (ICP4-siRNA) on mouse herpes simplex keratitis (HSK). Cy3-siRNA was mixed with Lipofectamine 2000, EntransterTM-in vivo, polyethyleneimine (PEI) or PEO-PPO-PEO pol...
