Human PLCG2 Haploinsufficiency Results in a Novel NK Cell Immunodeficiency
CONCLUSIONS: PLCG2 haploinsufficiency represents a distinct syndrome from previous variants characterized by NK cell immunodeficiency with herpes virus susceptibility, expanding the spectrum of PLCG2-related disease.PMID:37714437 | DOI:10.1016/j.jaci.2023.09.002

Dupilumab strengthens HSV-1 specific immune responses in atopic dermatitis
CONCLUSION: Our findings suggest that dupilumab may improve the HSV-1-specific immune response in AD due to an increased type I immune response and a reduction of HSV-1-specific IgE.PMID:37660986 | DOI:10.1016/j.jaci.2023.08.024

The role of food allergy as a provoking factor of migraine
CONCLUSION: We show for the first time the relationship between delayed-type food allergy and redistribution in the microbiome of the oropharynx of patients with migraine and once again confirm the role of delayed-type food allergy as a clinically significant factor influencing the course of migraine (its intensity and chronicity).PMID:37655411 | DOI:10.17116/jnevro202312308156

Dupilumab strengthens HSV-1 specific immune responses in atopic dermatitis
Impaired viral clearance in a subgroup of atopic dermatitis (AD) patients can lead to severe herpes simplex virus (HSV) infections named eczema herpeticum (EH). We recently identified a type 2 skewed viral immune response in EH patients. Clinical data suggest a reduced incidence of EH in AD patients treated with dupilumab, however, immunological investigations of this phenomenon are still lacking.

Comprehensive care of adults with respiratory diseases must include vaccines
Vaccines are among the most effective tools we have to improve and save lives, but only if they are administered to eligible patients. In this issue, Naeger et al1 highlight the major opportunities that the health care community has to achieve high adult vaccination rates. These vaccines include those for prevention of coronavirus disease 2019 (COVID-19), pneumococcal, and influenza, diseases that respiratory health professionals are most aware of, plus pertussis (in Tdap), herpes zoster (HZ, shingles), and as of May 3, 2023, respiratory syncytial viral, for which adults with asthma and chronic obstructive pulmonary diseas...

Dupilumab strengthens herpes simplex virus type 1 –specific immune responses in atopic dermatitis
Atopic dermatitis (AD), one of the most prevalent chronic inflammatory skin diseases, is orchestrated by a complex pathophysiology influenced by the immune system and skin barrier; it is influenced by environmental factors.1 Altered immune response is marked by type 2 inflammation, with a key role for TH2 cells, but it occurs especially in the chronic phase TH1 and TH17 responses as well.2,3 Moderately and severely affected AD patients are particularly prone to bacterial, fungal, and viral infections.

Stevens-johnson Syndrome and Toxic Epidermal Necrolysis: An Overview of Diagnosis, Therapy Options and Prognosis of Patients
Recent Adv Inflamm Allergy Drug Discov. 2023 Aug 21. doi: 10.2174/2772270817666230821102441. Online ahead of print.ABSTRACTBoth Stevens-Johnson syndrome (SJS) and Toxic-epidermal necrolysis (TEN) are generally medication-induced pathological conditions that mostly affect the epidermis and mucus membranes. Nearly 1 to 2 patients per 1,000,000 population are affected annually with SJS and TEN, and sometimes these maladies can cause serious life-threatening events. The reported death rates for SJS range from 1 to 5%, and 25 to 35% for TEN. The mortality risk may even be higher among elderly patients, especially in those who a...

Phenotypic and Endotypic Determinants of Atopic Dermatitis Severity from the ADRN Registry
CONCLUSION: In a diverse US AD population, severity was associated with a history of atopic disorders, skin and extracutaneous bacterial and viral infections (by history and physical examination), higher IgE, eosinophilia and allergen sensitization, atopic skin manifestations (i.e., excoriation, hyperlinear palms and ichthyosis), and atopic ocular features (i.e., blepharitis, conjunctivitis, and ectropion) as well as asthma findings (i.e., wheezing). Data from our prospective registry significantly advances our understanding of AD phenotypes and endotypes, which is critical to achieve optimal management.PMID:37182563 | DOI...

Severe Cutaneous Adverse Reaction Caused by Carbamazepine and Levofloxacin After Varicella Zoster Virus Infection
Infect Drug Resist. 2023 Mar 23;16:1705-1711. doi: 10.2147/IDR.S402267. eCollection 2023.ABSTRACTSevere cutaneous adverse reactions (SCARs) to drugs are associated with morbidity, mortality, healthcare costs, and challenges in drug development. It is important to identify the SCAR type early by using strict diagnostic criteria because they may require different treatments, follow-ups, and short- or long-term prognoses. A 68-year-old woman admitted to our hospital presented with fever and rashes for 10 days. This case exhibited many features that suggested acute generalized exanthematous pustulosis (AGEP). However, the cour...
