Serum Vitamin D and Risk of Breast Cancer within Five Years
Conclusions: In this cohort of women with elevated risk, high serum 25(OH)D levels and regular vitamin D supplement use were associated with lower rates of incident, postmenopausal breast cancer over 5 y of follow-up. These results may help to establish clinical benchmarks for 25(OH)D levels; in addition, they support the hypothesis that vitamin D supplementation is useful in breast cancer prevention. https://doi.org/10.1289/EHP943 About This Article Received: 09 August 2016 Revised: 23 January 2017 Accepted: 06 February 2017 Published: 06 July 2017 Address correspondence to C. From “Serum Sickness” to “Xenosialitis”: Past, Present, and Future Significance of the Non-human Sialic Acid Neu5Gc
Conclusions and Perspectives In this review, we have discussed important milestones from the early description of “Serum-sickness” as being due to antibodies directed against Neu5Gc epitopes all the way to the present-day therapeutic implications of these antibodies in cancer therapy. Some of these milestones have been represented in a concise timeline (Figure 6). While the “Xenosialitis” hypothesis is well-supported in the human-like mouse models, it has yet to be conclusively proven in humans. It remains to be seen if “Xenosialitis” plays a role in other uniquely-human dis...

Xenopus Resources: Transgenic, Inbred and Mutant Animals, Training Opportunities, and Web-Based Support
This reporter group also includes lines driven by ubiquitous promoters like CMV and human ubiquitin C. These are particularly useful for cut-and-paste, transplantation-based experiments to label and fate map regions of host embryos. The second group of reporter expression lines includes transgenics marking subcellular organelles (Supplementary Table S1B). These are highly useful for the study of molecular processes involved in cell function and are especially effective when utilized in the context of Xenopus egg extracts, the only cell-free system that permits full investigation of all DNA transactions related to cell cyc...

Isaac ’s story: A ‘new normal’ with short bowel syndrome
It was the morning after their baby son Isaac had come home from the hospital, and Jennifer and Brian Campbell were performing the same sweet act of bonding as many new parents: giving him a bath in the sink. But as they maneuvered around the room, they suddenly realized something was very wrong. “I fell to the floor and started screaming and crying as formula shot out of his stomach,” remembers Jennifer. “I thought we’d broken him.” The reality, of course, was that the Campbells were simply adjusting to their new “normal” — something any parent of a medically complex child can understand. Isaac wasn’t a ...

Fish oil for kidney transplant recipients.
CONCLUSIONS: There is insufficient evidence from currently available RCTs to recommend fish oil therapy to improve kidney function, rejection rates, patient survival or graft survival. The improvements in HDL cholesterol and diastolic blood pressure were too modest to recommend routine use. To determine a benefit in clinical outcomes, future RCTs will need to be adequately powered with these outcomes in mind. PMID: 27535773 [PubMed - as supplied by publisher]

Presentation Pattern, Diagnostic Markers, Management and Outcome of IgD Multiple Myeloma: A Systematic Review
Conclusion:N-glycan assay can overcome the limitations of SPEP and quantitative serum IgD assay in the diagnosis of IgD MM. This difficulty is posed by the lower concentrations of M spike ( SPEP) or lower-normal serum IgD value on quantitative serum assays, which can delay its diagnosis. Novel and conventional agents have good responses with ORR>90%, but mOS of conventional agents was lower when compared to novel agents. Bortezomib based regimens showed better responses when compared to non-bortezomib regimens. The best response (ORR=95%) was obtained with use of SCT in combination with novel agents. Serum free light ch...

Straight from the heart: Mysterious lipids may predict cardiac problems better than cholesterol
Stephanie Blendermann, 65, had good reason to worry about heart disease. Three of her sisters died in their 40s or early 50s from heart attacks, and her father needed surgery to bypass clogged arteries. She also suffered from an autoimmune disorder that results in chronic inflammation and boosts the odds of developing cardiovascular illnesses. “I have an interesting medical chart,” says Blendermann, a real estate agent in Prior Lake, Minnesota. Yet Blendermann’s routine lab results weren’t alarming. At checkups, her low-density lipoprotein (LDL), or “bad,” cholesterol hovered around the 100 milligrams-per-...

Bifunctional Small Molecules Enhance Neutrophil Activities Against Aspergillus fumigatus in vivo and in vitro
We report that the bifunctional compounds enhance the interactions between primary human neutrophils and A. fumigatus in vitro, using three microfluidic assay platforms. The bifunctional compounds significantly enhance the recruitment of neutrophils, increase hyphae killing by neutrophils in a uniform concentration of drug, and decrease hyphal tip growth velocity in the presence of neutrophils compared to the antifungal targeting moiety alone. We validated that the bifunctional compounds are also effective in vivo, using a zebrafish infection model with neutrophils expressing the appropriate EM receptor. We measured signif...

Effects of Neurotrophic Factors in Glial Cells in the Central Nervous System: Expression and Properties in Neurodegeneration and Injury
Conclusion and Future Aspects This review summarizes available NTF expression data, compiles existing evidence on the effects of glial NTF signaling in healthy conditions and in disease models (Figure 1), and highlights the importance of this topic for future studies. The relationship between NTFs and glia is crucial for both the developing and adult brain. While some of these factors, such as NT-3 and CNTF, have highly potent effects on gliogenesis, others like BDNF and GDNF, are important for glia-mediated synapse formation. Neurotrophic factors play significant roles during neurodegenerative disorders. In many cases, ...

How to deep freeze an entire organ —and bring it back to life
MINNEAPOLIS— The rat kidney on the operating table in front of Joseph Sushil Rao looked like it had been through hell. Which it had—a very cold one. Normally a deep pink, this thumbnail-size organ was blanched a corpselike gray. In the past 6 hours, it had been plucked from the abdomen of a white lab rat, pumped full of a black fluid, stuck in a freezer cooled to –150°C, and zapped by a powerful magnet. Now, in a cramped, windowless room on the 11th floor of the University of Minnesota’s (UMN’s) Malcolm Moos Health Sciences Tower, Rao lifted the kidney from a small plastic box and gently laid it ins...

Outcomes of Patients with Relapsed/Refractory Double Expressor B Cell Lymphoma As Defined By Multicenter Pathology Review Treated with Ibrutinib Monotherapy
Conclusion: An objective response to ibr was experienced by nearly half of pts with R/R DEL as defined by multicenter HP review in this series. Our data suggest a potential benefit to the use of ibr monotherapy as bridging tx to curative-intent cellular tx, as well as support the incorporation of ibr into clinical trials for the R/R DEL pt population. A relatively high rate of concordance between HP1 and CPR in identifying cases of DEL suggests that local review of MYC and BCL2 IHC may be a reasonable alternative for determining DEL status when central review is infeasible.DisclosuresLandsburg: Curis, INC: Consultancy, Res...

MRD-Response Driven Phase I/II Study for Newly Diagnosed Multiple Myeloma Patients Using Higher Doses of Twice-Weekly Carfilzomib (45 and 56 mg/m2) in Combination with Lenalidomide and Dexamethasone
ConclusionIn this phase I/II clinical trial assessing higher doses of twice-weekly Car dosing in combination with Len and Dex, we established MTD 56 mg/m2 and demonstrated a MRD platform using multi-parametric flow cytometry can be successfully used to tailor individualized treatment plans. Higher doses of twice weekly Car (45 and 56 mg/m2) in combination with Len and Dex, resulted in rapid and deep responses with approximately 60% MRD negative rate, and a safety profile similar to KRD standard dose (Car 36 mg/m2).Table.DisclosuresKorde: Amgen: Research Funding.
