Development of Injectable Thermosensitive Nanocomposite Hydrogel for Ratiometric Drug Delivery to Treat Drug Resistant Chondrosarcoma In Vivo
A radiometric nanocomposite hydrogel capable of co-delivering Doxorubicin and the Bcl-2 inhibitor ABT-737 is engineered, employing an optimized drug ratio of 5:1 (w/w) suitable for both in vitro and in vivo applications. The findings reveals a synergistic tumor inhibition effect with the co-delivery hydrogel. This innovative approach presents a transformative strategy for addressing drug resistance in chondrosarcoma chemotherapy, holding substantial promise for clinical applications. AbstractChondrosarcoma(CS), a prevalent primary malignant bone tumor, frequently exhibits chemotherapy resistance attributed to upregulated a...

Research progress on the PEGylation of therapeutic proteins and peptides (TPPs)
With the rapid advancement of genetic and protein engineering, proteins and peptides have emerged as promising drug molecules for therapeutic applications. Consequently, there has been a growing interest in the field of chemical modification technology to address challenges associated with their clinical use, including rapid clearance from circulation, immunogenicity, physical and chemical instabilities (such as aggregation, adsorption, deamination, clipping, oxidation, etc.), and enzymatic degradation. Polyethylene glycol (PEG) modification offers an effective solution to these issues due to its favorable properties. This...

Molecules, Vol. 29, Pages 1217: Responsive Microgels through RAFT-HDA Dynamic Covalent Bonding Chemistry
uo Jun Ji This paper developed a method for preparing ultrasound-responsive microgels based on reversible addition fragmentation chain transfer-hetero Diels–Alder (RAFT-HAD) dynamic covalent bonding. First, a styrene cross-linked network was successfully prepared by a Diels–Alder (DA) reaction between phosphoryl dithioester and furan using double-ended diethoxyphosphoryl dithiocarbonate (BDEPDF) for RAFT reagent-mediated styrene (St) polymerization, with a double-ended dienophile linker and copolymer of furfuryl methacrylate (FMA) and St as the dienophile. Subsequently, the microgel system...

Tailoring Zinc Ferrite Nanoparticle Surface Coating for Macrophage-Affinity Magnetic Resonance Imaging of Atherosclerosis
ACS Appl Mater Interfaces. 2024 Mar 6. doi: 10.1021/acsami.3c17212. Online ahead of print.ABSTRACTAtherosclerosis is a chronic inflammatory disease characterized by the formation of atherosclerotic plaques, while macrophages as key players in plaque progression and destabilization are promising targets for atherosclerotic plaque imaging. Contrast-enhanced magnetic resonance imaging (CE-MRI) has emerged as a powerful noninvasive imaging technique for the evaluation of atherosclerotic plaques within arterial walls. However, the visualization of macrophages within atherosclerotic plaques presents considerable challenges due t...

Glycerol- and diglycerol-based polyesters: Evaluation of backbone alterations upon nano-formulation performance
Colloids Surf B Biointerfaces. 2024 Feb 28;236:113828. doi: 10.1016/j.colsurfb.2024.113828. Online ahead of print.ABSTRACTDespite the success of polyethylene glycol-based (PEGylated) polyesters in the drug delivery and biomedical fields, concerns have arisen regarding PEG's immunogenicity and limited biodegradability. In addition, inherent limitations, including limited chemical handles as well as highly hydrophobic nature, can restrict their effectiveness in physiological conditions of the polyester counterpart. To address these matters, an increasing amount of research has been focused towards identifying alternatives to...

Real-world effectiveness of GLP-1 receptor agonist-based treatment strategies on “time in range” in patients with type 2 diabetes
Conclusion: These real-world findings indicate that GLP-1RA-based treatment strategies could be superior to oral treatment strategies for improving TIR among patients with T2DM and that once-weekly GLP-1RA may be more effective than a once-daily GLP-1RA.Clinical trial registration:http://www.chinadrugtrials.org.cn/index.html, identifier number ChiCTR2300073697. Thermally Reversible Gel-Sol Transition of Hydrogels via Dissociation and Association of an Artificial Protein Nanocage
Biomacromolecules. 2024 Mar 6. doi: 10.1021/acs.biomac.3c01285. Online ahead of print.ABSTRACTOligomeric protein nanocages often disassemble into their subunits and reassemble by external stimuli. Thus, using these nanocages as cross-linkers for hydrogel network structures is a promising approach to allow hydrogels to undergo stimuli-responsive gel-sol transitions or self-healing. Here, we report hydrogels that show a reversible gel-sol transition resulting from the heat-induced dissociation and reassociation of protein nanocages. The hydrogel contained the 60-mer artificial protein nanocage, TIP60, as a supramolecular cro...
