Carbon chain length of perfluoroalkylated carboxylic acids determines inhibitory strength on gonadal 3 β-hydroxysteroid dehydrogenases in humans, rats, and mice
In conclusion, the carbon chain length determines the inhibitory strength of PFCAs on human, rat, and mouse gonadal 3β-HSDs, and the inhibitory strength of PFCAs against human and rat 3β-HSD enzymes shows V-shaped turn.PMID:37871704 | DOI:10.1016/j.toxlet.2023.10.011 (Source: Toxicology Letters)

Early-life ozone exposure modulates region-specific gene expression in the developing rat lung
This study aimed to elucidate changes in gene expression following episodic ozone exposure to identify potential mechanisms of ozone-mediated impairments in lung development. Rat pups were exposed to either filtered air or ozone (0.5 ppm, 6hr./day, 5 days/week) from postnatal day (PND) 7-28 (16 dams total with 8 pups each, 4M & 4F) and sacrificed at either PND 30-31 or PND 80-84. Lung microdissection isolated major regions for RNA-Seq analysis. Ozone modified inherent differences in gene expression between lung regions in both male and female rat pups, whereas statistically significant changes in gene expression direct...

Determination of the elimination half-life of Glyphosate and its main metabolite, AMPA, in chicken plasma
In this study, toxicokinetic parameters for glyphosate and AMPA were calculated after one unique dietary exposure (40 mg of glyphosate equivalent per kg) and one unique intravenous injection of a GBH, in hens and roosters respectively. Non compartmental analysis was used to show the evolution of glyphosate and AMPA plasma concentrations over time. After one unique intravenous injection of a glyphosate-based herbicide, glyphosate and AMPA were quickly eliminated from plasma and were poorly distributed (Vssglyphosate = 0.30 L/kg). Their terminal half-lives are 4.7 h and 8.10 h, respectively. After dietary exposure, glyphosat...

Dichotomous effect of methylphenidate on microglia and astrocytes: Insights from in vitro and animal studies
Toxicol Lett. 2023 Oct 14;389:1-10. doi: 10.1016/j.toxlet.2023.10.008. Online ahead of print.ABSTRACTMethylphenidate (MPH) has been used for decades to treat attention-deficit/hyperactivity disorder (ADHD) and narcolepsy. Moreover, several studies have shown that it is subject to misuse, particularly among college students and adolescents, for cognitive enhancement or as a recreational drug. This phenomenon causes concern, and it is critical to clarify better how MPH impacts brain cells. In fact, data has suggested that MPH could result in neuroinflammation and neurodegeneration across several brain regions; however, littl...

The suitability of a polydimethylsiloxane-based (PDMS) microfluidic two compartment system for the toxicokinetic analysis of organophosphorus compounds
Toxicol Lett. 2023 Oct 10;388:24-29. doi: 10.1016/j.toxlet.2023.10.007. Online ahead of print.ABSTRACTOrgan-on-a-chip platforms are an emerging technology in experimental and regulatory toxicology (species-specific differences, ethical considerations). They address gaps between in vivo and in vitro models. However, there are still certain limitations considering material, setup and applicability. The current study examined the suitability of a commercially available polydimethylsiloxane-based (PDMS) organ-chip for the toxicokinetic characterization of the highly toxic nerve agent VX and the organophosphate pesticide parath...
