Cradle cultures: growing stem cell-derived developmental cell models < em > in vitro < /em >
Biotechniques. 2023 Nov 16. doi: 10.2144/btn-2023-0100. Online ahead of print.ABSTRACTHow are three stem cell-derived developmental cell models furthering our understanding of post-implantation human embryo development? And why have recent advancements in these human embryo-like models spurred ethical discussion and the need to refine our definition of 'embryo'? Predicting molecular docking of per- and polyfluoroalkyl substances  to blood protein using generative artificial intelligence algorithm DiffDock
This study computationally evaluates the molecular docking affinity of various perfluoroalkyl and polyfluoroalkyl substances (PFAs) towards blood proteins using a generative machine-learning algorithm, DiffDock, specialized in protein-ligand blind-docking learning and prediction. Concerns about the chemical pathways and accumulation of PFAs in the environment and eventually in the human body has been rising due to empirical findings that levels of PFAs in human blood has been rising. DiffDock may offer a fast approach in determining the fate and potential molecular pathways of PFAs in human body.PMID:37947020 | DOI:10.2144...

Considerations for colorblind individuals on selecting colorimetric or fluorescent dye assay outcomes
Biotechniques. 2023 Nov 9. doi: 10.2144/btn-2023-0041. Online ahead of print.ABSTRACTA disadvantage of colorimetric detection in nucleic acid amplification assays is the possibility that a colorblind individual may interpret colors differently than observers with full-color vision. Using an isothermal amplification assay, the ability of colorblind individuals to distinguish between positive and negative results for four dyes was tested. Five individuals with self-reported colorblindness and four with full-color vision reported their observations of the color of the solution. Although colorblind individuals may accurately i...

Simple, fast and inexpensive hot sodium hydroxide and tris DNA extraction method for genotyping tomato and melon seeds
Biotechniques. 2023 Nov 2. doi: 10.2144/btn-2023-0054. Online ahead of print.ABSTRACTSeed commerce is a highly profitable global market. Most commercialized seeds are hybrid seeds originating from a controlled cross between two selected parental lines. The market value of hybrid seeds depends on their hybrid genetic purity. DNA molecular markers are a reliable and widespread tool to genotype plant materials; however, DNA extraction from seeds is challenging, often laborious and expensive. With the ultimate goal of creating a tomato and melon hybrid seeds purity test, various challenges arise. To overcome these problems and...

MSCProfiler: a single cell image processing workflow to investigate mesenchymal stem cell heterogeneity
In this study, the authors describe MSCProfiler, an image analysis pipeline developed using CellProfiler software. This workflow can extract geometrical and texture features such as shape, size, eccentricity and entropy, along with intensity values of the surface markers from multiple single cell images obtained using imaging flow cytometry. This screening pipeline can be used to analyze geometrical and texture features of all types of MSCs across different passages hallmarked by enhanced feature extraction potential from brightfield and fluorescent images of the cells.PMID:37916466 | DOI:10.2144/btn-2023-0048 (Source: BioTechniques)
