A recalcitrant skin lesion and subsequent infection in a recreational intramural male athlete: a case report
J Athl Train. 2023 Nov 28. doi: 10.4085/1062-6050-0467.22. Online ahead of print.ABSTRACTA 35-year-old intramural male athlete presented to the athletic training staff with a 4.5cm x 2.2cm itchy, painful, swollen, and infected insidious skin lesion on his right lateral malleolus due to an underlying dermatological deficiency. Suspecting infection, the patient was referred to his nurse practitioner and was diagnosed with atopic dermatitis caused by a ceramide deficiency. He was placed on Cefalexin and Mupirocin 2% ointment but returned due to the lesion increasing to 8.5cm x 6cm although infection seemed controlled. He was ...

Unseen steroids, seen risks: Skin-Cap still contains high-potency glucocorticosteroids 26  years after global scandal
To the Editor: Skin-Cap spray (formerly Cheminova Laboratories International; now Chemigroup France) is an over-the-counter medication for psoriasis and other dermatoses. According to the manufacturer's statement, the active ingredient is zinc pyrithione. However, from 1997 to 2005, the presence of high-potency glucocorticosteroids, specifically clobetasol propionate and betamethasone propionate, was detected in Skin-Cap's composition.1,2 This evidence prompted the recall of Skin-Cap in the United States, Canada, and several European countries. Divergent Chemo- and Biocatalytic Route to 16 β-Methylcorticoids: Asymmetric Synthesis of Betamethasone Dipropionate, Clobetasol Propionate, and Beclomethasone Dipropionate
Angew Chem Int Ed Engl. 2023 Nov 22:e202313952. doi: 10.1002/anie.202313952. Online ahead of print.ABSTRACT16β-Methylcorticoids are among the most important glucocorticoid steroids for the treatment of various dermatological disorders, respiratory infections, and other allergic reactions elicited during inflammatory responses of the human body. Betamethasone dipropionate, clobetasol propionate, and beclomethasone dipropionate are particularly noteworthy for their synthetic intractability. Despite five decades of research, these 16β-methylcorticoids have remained challenging synthetic targets owing to insurmountable issue...

Anti-laminin 332 antibodies in graft-versus-host disease-associated bullous pemphigoid after allogeneic peripheral blood stem cell transplantation
We report a 48-year-old woman with bullous pemphigoid (BP) with antibodies against the B3 subunit of laminin 332 after the development of graft-versus-host disease (GVHD). She was diagnosed with recurrent acute lymphoblastic leukemia at 40 years of age and underwent two rounds of allogeneic peripheral blood stem cell transplantations (PBST). Two and a half years after the second PBST, multiple tense blisters appeared on her face, hands, and lower legs. The diagnosis of BP was based on hematoxylin eosin and immunofluorescence staining and immunoblotting analyses. A combination regimen of topical corticosteroids (clobetasol ...

Transcriptomic profiling of TLR-7-mediated immune-challenge in zebrafish embryos in the presence and absence of glucocorticoid-induced immunosuppression
Ecotoxicol Environ Saf. 2023 Oct 14;266:115570. doi: 10.1016/j.ecoenv.2023.115570. Online ahead of print.ABSTRACTAlthough numerous studies imply a correlation between chemical contamination and an impaired immunocompetence of wildlife populations, the assessment of immunomodulatory modes of action is currently not covered in the regulatory requirements for the approval of new substances. This is not least due to the complexity of the immune system and a lack of standardised methods and validated biomarkers. To tackle this issue, in this study, the transcriptomic profiles of zebrafish embryos were analysed in response to th...

Management of Pediatric Psoriasis: A U.S. Survey Based on Visits from the National Ambulatory Medical Care Survey (NAMCS)
ConclusionUse of off-label treatments was common for pediatric psoriasis. Most children with psoriasis were treated with topicals, of which tacrolimus, an unapproved treatment, was the most common. The frequent use of tacrolimus could indicate an avoidance of corticosteroids in children. (Source: Dermatology and Therapy)

Molecules, Vol. 28, Pages 6907: Cannabidiol-Loaded Lipid-Stabilized Nanoparticles Alleviate Psoriasis Severity in Mice: A New Approach for Improved Topical Drug Delivery
C. Sintov Cannabidiol (CBD) is a promising natural agent for treating psoriasis. CBD activity is attributed to inhibition of NF-kB, IL-1β, IL-6, and IL-17A. The present study evaluated the anti-psoriatic effect of cannabidiol in lipid-stabilized nanoparticles (LSNs) using an imiquimod (IMQ)-induced psoriasis model in mice. CBD-loaded LSNs were stabilized with three types of lipids, Cetyl alcohol (CA), Lauric acid (LA), and stearic-lauric acids (SALA), and were examined in-vitro using rat skin and in-vivo using the IMQ-model. LSNs loaded with coumarin-6 showed a localized penetration depth of about 100 &...
