A sensitive and efficient LC-MS/MS method for the bioanalysis of Fosinopril diacid from human plasma and its application for a bioequivalence study in humans.
Abstract Fosinopril diacid is an ACE inhibitor with efficient anti-hypertensive action. It is the active metabolic product formed in the body on hydrolysis of its prodrug Fosinopril. A sensitive, rapid method with high recovery for Fosinopril diacid from human plasma was developed. Solid phase extraction technique employing Waters Oasis SPE cartridges gave clean samples with very high recovery of 97%. The analyte along with its internal standard (Benazepril hydrochloride) were chromatographed on XTerra RP8 column (4.6 x 50 mm, 5μm) using Methanol- Ammonium acetate buffer (10 mM) (90:10, v/v) as the mobile...

Cardiovascular Benefits of Angiotensin Converting Enzyme Inhibition plus Calcium Channel Blockade in Patients Achieving Tight Blood Pressure Control and with Resistant Hypertension.
CONCLUSIONS: Combination therapy adding a calcium channel blocker, rather than hydrochlorothiazide, to an angiotensin converting enzyme inhibitor was more effective in preventing composite cardiovascular events even in hypertensive patients achieving aggressive systolic blood pressure targets as well as in those with apparent resistant hypertension. Our findings add support that most patients, including those following contemporary clinical guidelines, will benefit from this combination. TRIAL REGISTRATION: ClinicalTrials.gov, NCT00170950. Chrysophanol Protects Against Acute Heart Failure by Inhibiting JNK1/2 Pathway in Rats.
CONCLUSIONS Chrysophanol improved cardiac functions and protected against myocardial damage, apoptosis, OS, and inflammation by inhibiting activation of the JNK1/2 pathway in AHF rat models. These finding indicate that chrysophanol may be a promising approach for treatment of AHF. PMID: 33044948 [PubMed - in process] (Source: Medical Science Monitor)

Safety implications of combining ACE inhibitors with thiazides for the treatment of hypertensive patients.
Authors: Borghi C, Soldati M, Bragagni A, Cicero AFG Abstract INTRODUCTION: ACE-inhibitors (ACEI) and diuretics are the typical first-line antihypertensive drugs with complementary mechanisms of action. The present paper is summarizing the evidence supporting the efficacy of their combination in a broad range of hypertensive patients. AREAS COVERED: This source of data are different trials investigating the use of ACEI and diuretics in different populations of patients. The available evidence supports some advantage for thiazide-type compounds (chlortalidone-CHT and indapamide-IND) in the preventio...

Postnatal renin-angiotensin system inhibition prevents renal damage from prenatal inflammation in rats.
CONCLUSION: Together with our previous findings that postnatal inhibition of nuclear factor-kappa B activation blocks intra-renal renin-angiotensin system activation, our current data demonstrate that intra-renal activation of the renin-angiotensin system interacts with nuclear factor-kappa B activation to cause renal damage in adulthood following prenatal inflammation. DELay of appearance of sYmptoms of canine degenerative mitral valve disease treated with spironolactone and benazepril: The DELAY study
ConclusionsThis study failed in demonstrating that combined administration of spironolactone and benazepril delays onset of HF in dogs with preclinical MMVD. However, such treatment induces beneficial effects on cardiac remodeling and these results could be of clinical relevance. (Source: Journal of Veterinary Cardiology)
