GSE175671 U2AF1 plays a critical role in global co-transcriptional splicing (Fraction chrRNA-seq)
Contributors : Yajing Hao ; Changwei Shao ; Xiang-Dong FuSeries Type : Expression profiling by high throughput sequencingOrganism : Homo sapiensIn order to measure the impact of U2AF1 on co-transcriptional splicing, we treated HeLa cells with control siRNA or U2AF1 siRNA. To measure co-transcriptional splicing, we fractionated HeLa cells into cytosolic (Cyt), nucleoplasm (NP), and chromatin/particle (Chr) fractions. We purified total RNA from Chr compartment, spiked in with in vitro transcribed EGFP and FireFly luciferase RNA for data normalization, depleted ribosomal RNAs, and constructed two independent libraries for RNA-seq.

Genes, Vol. 14, Pages 1157: High Level of GMFG Correlated to Poor Clinical Outcome and Promoted Cell Migration and Invasion through EMT Pathway in Triple-Negative Breast Cancer
In this study, data from the Cancer Genome Atlas (TCGA), Clinical Proteomic Tumor Analysis Consortium (CPTAC), Human Protein Atlas (HPA), and Genotype-Tissue Expression (GTEx) databases were used to analyze the expression of GMFG in pan-cancer and the correlation between clinical factors. Gene Set Cancer Analysis (GSCA) and Gene Set Enrichment Analysis (GSEA) were also used to analyze the functional differences between the different expression levels and predict the downstream pathways. GMFG expression in breast cancer tissues, and its related biological functions, were further analyzed by immunohistochemistry (IHC), immun...

GSE209768 Gene expression in AML12 cells after Uhrf2 knock-down
Contributors : Coenraad F Slabber ; Andrii Kuklin ; Sabine WernerSeries Type : Expression profiling by high throughput sequencingOrganism : Mus musculusWe identified the ubiquitin ligase Uhrf2 as a key regulator of liver regeneration. To determine the underlying mechanisms of action, we performed siRNA-mediated knockdown of Uhrf2 in AML12 cells and analysed the transcriptome of the Uhrf2-deficient cells.

GSE232483 Effect of SRCAP on gene expression during PRRSV infection of Marc-145 cells.
Contributors : Yihong Xiao ; Guofei DingSeries Type : Expression profiling by high throughput sequencingOrganism : Chlorocebus sabaeusTo investigate the SRCAP regulation of gene transcription during PRRSV infection, we established Marc-145 cell lines in which SRCAP gene has been knocked down by siRNA.We then performed gene expression profiling analysis using data obtained from RNA-seq.

GSE227774 Differential gene expression profile analysis of parental, Cas9-p15 control, and MELK knockout cells
Contributor : Chandra BartholomeuszSeries Type : Expression profiling by arrayOrganism : Homo sapiensTriple-negative breast cancer (TNBC) has high relapse and metastasis rates and a high proportion of cancer stem-like cells (CSCs), which possess self-renewal and tumor initiation capacity. MELK (maternal embryonic leucine zipper kinase), a protein kinase of the Snf1/AMPK kinase family, is known to promote CSC maintenance and malignant transformation. Our study showed that MELK knockdown using siRNA or MELK inhibition using the MELK inhibitor MELK-In-17 significantly reduced invasiveness, reversed epithelial-to-mesenchymal t...
