Case Report: Lack of Response to Givosiran in a Case of ALAD Porphyria
Discussion: No adverse effects of givosiran on the liver were observed in this patient with cirrhosis during 6 months of treatment with givosiran. The patient has continued to have recurrent attacks, with transient decreases in ALA levels only as related to treatment of his attacks with hemin. Our experience limited to one patient with ADP suggests that givosiran may not be effective in this type of acute porphyria. Since ADP may have an erythropoietic component, treatment with hydroxyurea, which was beneficial in one previous case, is planned.

GSE161899 The dependency factor RRM2 at the nexus of a copy number driven regulatory network and a target for synthetic lethal interactions with replication stress checkpoint addiction in high-risk neuroblastoma siRNA_RRM2
In this study, we identified and functionally evaluated the ‘ribonucleotide reductase regulatory subunit 2’ (RRM2) as a top-ranked 2p putative co-driver and therapeutic target in high-risk neuroblastoma enforcing replicative stress resistance. RRM2 dependency was shown in vitro through knockdown in neuroblastoma cells, with transcriptome analysis reveali ng downregulation of pediatric tumor markers. Transgenic zebrafish lines expressing RRM2 in immature sympathoblasts were crossed with a dh-MYCN transgenic zebrafish line, with RRM2 overexpression dramatically increasing tumor penetrance from 20% to nearly 100%, acce...

GSE210017 NAT10 modulates fatty acid metabolism of palmitate driven MCF7 in ac4C dependent manner
Contributors : Dalhat M Hassan ; Khan Mohammad ImranSeries Type : Expression profiling by high throughput sequencingOrganism : Homo sapiensWe show that non-toxic exogenous palmitate uptake in MCF7 cells promotes NAT10 expression and NAT10-dependent ac4C RNA modification. It was previously reported that NAT10 modulates the addition of ac4C on RNA transcripts in normal and cancer conditions. However, no study report the impact of NAT10 in palmitate driven cells. Here we performed RNA immunoprecipitation sequencing (RIP-seq) in palmitate loaded MCF7 knockdown with NAT10 siRNA. Based on the pathways and enrichment landscape id...

GSE189458 The RNA sequencing analysis of nasopharyngeal carcinoma (NPC) cell line S18 upon circIPO7 knockdown
Contributors : X H Hong ; J Ma ; Y Q LiSeries Type : Expression profiling by high throughput sequencingOrganism : Homo sapiensThe development of therapeutic resistance and metastatic dissemination takes place in 20-30% patients with nasopharyngeal carcinoma (NPC), resulting in poor survival. Hence, a better understanding of the underlying molecular mechanisms will help improve clinical outcome. We have identified a novel circRNA, circIPO7, as a promoter of metastasis and cisplatin chemoresistance in NPC cells. High-throughput RNA sequencing analysis was performed in the human nasopharyngeal carcinoma cell line S18 after tr...

GSE185932 Gene expression changes associated with reducing Sox2 expression in ovarian cancer cell line PA1 under anchorage independence
The objective of this study is to examine the effect of changing Sox2 expression under the conditions of anchorage independent growth that is known to drive the expression of survival genes. We compared genome wide gene expression profiles of the ovarian tumor cell line PA1 upon transient knockdown of Sox2 expression using siRNA and compared to negative silencer control siRNA cells. Cells grown in biological triplicates were transfected under steady state 2D growth conditions and then transferred to poly-hema coated plates to prevent attachment. After 72 hrs, RNA was collected from siSox2 and sicontrol cells followed by ve...

GSE183793 Global gene expression of canine bladder K9TCC cells and cancer stem cells after COX-2 inhibition
Contributors : Emma A Hurst ; Lisa Y Pang ; David J ArgyleSeries Type : Expression profiling by arrayOrganism : Canis lupus ; Canis lupus familiarisCancer stem cells (CSCs) are fundamental to bladder cancer progression and the cyclooxygenase-2 (COX-2) pathway has a pro-tumorigenic role in CSC function. Naturally occurring bladder cancer in dogs has the potential to be a natural model of the human disease. Here, we used canine specific microarrays to determine changes in global gene expression of canine bladder cancer cells (the K9TCC cell line) and bladder cancer stem cells isolated from the K9TCC cell line after treatment...

GSE207658 Functional analysis across model systems implicates ribosomal proteins in growth and proliferation defects associated with hypoplastic left heart syndrome
Contributors : Georg Vogler ; Alexandre R ColasSeries Type : Expression profiling by high throughput sequencingOrganism : Homo sapiensHypoplastic left heart syndrome (HLHS) is the most lethal congenital heart disease (CHD). The pathogenesis of HLHS is poorly understood and due to the likely oligogenic complexity of the disease, definitive HLHS-causing genes have not yet been identified. Postulating that impaired cardiomyocyte proliferation as a likely important contributing mechanism to HLHS pathogenesis, and we conducted a genome-wide siRNA screen to identify genes affecting proliferation of human iPSC-derived cardiomyocy...

Intelligent Nanoparticles With pH-Sensitive Co-Delivery of Temozolomide and siEGFR to Ameliorate Glioma Therapy
In this study, pH-sensitive and GBM-targeting nanovesicle (Tf-PEG-PAE(SS)) was fabricated. The chemotherapy drug (TMZ) and EGFR inhibitor (EGFR-siRNA) were co-encapsulated in the nanocarrier, and their anticancer outcomes were investigated in detail. In vitro experiments have shown that the nanocarrier transports TMZ and EGFR-siRNA efficiently into U87 cells, causing a vigorous apoptotic response by silencing the proliferative EGFR gene and increasing the drug concentration of TMZ simultaneously. An experimental study in mice bearing orthotropic glioma revealed that the accumulated nanocarriers in the tumor site could inhi...

Deletion of microRNA-183-96-182 Cluster in Lymphocytes Suppresses Anti-DsDNA Autoantibody Production and IgG Deposition in the Kidneys in C57BL/6-Faslpr/lpr Mice
This study reports that in vitro inhibition of miR-182 alone or miR-183C by specific antagomirs in activated splenocytes from autoimmune-prone MRL/lpr and control MRL mice significantly reduced lupus-related inflammatory cytokines, interferon-gamma (IFNγ), and IL-6 production. To further characterize the role of miR-182 and miR-183C cluster in vivo in lupus-like disease and lymphocyte phenotypes, we used hCD2-iCre to generate B6/lpr mice with conditional deletion of miR-182 or miR-183C in CD2+ lymphocytes (miR-182−/−B6/lpr and miR-183C−/-B6/lpr). The miR-182−/−B6/lpr and miR-183C−/−B6/lpr mice had significan...
