Ursodesoxycholic acid is an FFA4 agonist and reduces hepatic steatosis via FFA4 signaling
This study not only identified a new skeleton of FFA4 agonists, but also demonstrated that FFA4 signal was accounting for the protective effects of UDCA in the NAFLD treatment.PMID:35033554 | DOI:10.1016/j.ejphar.2022.174760

Piezo1 mediates endothelial atherogenic inflammatory responses via regulation of YAP/TAZ activation
AbstractThe vascular endothelium plays a key role in the pathobiology of atherosclerotic cardiovascular disease. Endothelial cell Piezo1 mediates blood vessel formation, angiogenesis and regulation of blood pressure. However, changes of Piezo1 expression in atherosclerosis (AS) and the role of Piezo1 in the progression of atherosclerotic diseases remains obscure. Thus, the current study is to elucidate the role and mechanism of which Piezo1 mediates vascular inflammation in atherosclerotic mice and vascular endothelial inflammation induced by oxidized low density lipoprotein (ox-LDL) in vitro. Here, we have shown that the ...

Anti-inflammatory and anti-fibrotic effects of modafinil in nonalcoholic liver disease
Biomed Pharmacother. 2021 Dec;144:112372. doi: 10.1016/j.biopha.2021.112372. Epub 2021 Oct 28.ABSTRACTSmall- and intermediate-conductance Ca2+-activated K+ channels, KCa2.3 and KCa3.1, are involved in cellular signaling processes associated with inflammation and fibrosis. KCa2.3 and KCa3.1 are upregulated by proinflammatory cytokines and profibrotic growth factors. Cyclic AMP, which downregulates KCa2.3 and KCa3.1, is elevated by modafinil in cells; accordingly, we investigated whether modafinil exerts anti-inflammatory and anti-fibrotic responses via KCa2.3- and KCa3.1-mediated pathways in high-fat diet (HFD)- or thioacet...

Carnosol alleviates nonalcoholic fatty liver disease by inhibiting mitochondrial dysfunction and apoptosis through targeting of PRDX3
In conclusion, CAR suppressed lipid accumulation, mitochondrial dysfunction and hepatocyte apoptosis by activating PRDX3, mitigating the progression of NAFLD, and thus, CAR may represent a promising candidate for clinical treatment of steatosis.PMID:34678374 | DOI:10.1016/j.taap.2021.115758

MMP-12 siRNA improves the homeostasis of the small intestine and metabolic dysfunction in high-fat diet feeding-induced obese mice
Biomaterials. 2021 Oct 11;278:121183. doi: 10.1016/j.biomaterials.2021.121183. Online ahead of print.ABSTRACTThe changes of small intestinal homeostasis have been recognized to contribute essentially to the obese development. However, the core small intestinal regulator which mediates over-nutrient impacts on the homeostasis of the small intestines remains elusive. Here, we identify the MMP-12 as such a responsive factor in mouse small intestines. Taking advantages of the nano delivery system, we demonstrate that small intestine-specific MMP-12 knockdown alleviates high-fat diet feeding-induced metabolic disorders and impr...

MiR-124 Prevents the Microglial Proinflammatory Response by Inhibiting the Activities of TLR4 and Downstream NLRP3 in Palmitic Acid-Treated BV2 Cells
AbstractNeuroinflammation is a mechanism by which obesity or a high-fat diet leads to cognitive impairment. MiR-124, a highly expressed microRNA in the brain, can alleviate neuroinflammation by regulating microglial activation, but its mechanism is unclear. The aim of the study was to explore whether miR-124 exerted this effect through TLR4/MyD88/NF- κB p65/NLRP3 signaling in palmitic acid-treated BV2 cells. Prepared BV2 cells were treated with palmitic acid to establish an in vitro model of a high-fat diet. An miR-124 mimic and inhibitor were adopted to upregulate and downregulate the expression of miR-124, respectively....

High-fat diet exacerbated BDE-209-induced hepatocyte apoptosis via intensifying the transfer of Ca < sup > 2+ < /sup > from endoplasmic reticulum to mitochondria
This study aimed to evaluate the hepatotoxicity of BDE-209 in lean and high-fat diet (HFD)-treated obese mice and elucidate the underlying mechanisms. Firstly, the increasing levels of triacylglycerol and proinflammatory factors in the liver and ALT and AST in serum demonstrated the hepatic damage caused by BDE-209 and further exacerbated by HFD. Tunel image revealed that BDE-209 induced more severe hepatocyte apoptosis with the assistant of HFD. Next, the mechanism analysis showed that the pro-apoptotic action of BDE-209 was in an endoplasmic reticulum (ER)/Ca2+ flux/mitochondria-dependent manner, concluded from the impai...

Morin hydrate inhibits atherosclerosis and LPS-induced endothelial cells inflammatory responses by modulating the NF κB signaling-mediated autophagy
In conclusion, MO inhibits atherosclerosis in ApoE-/- mice and LPS-induced inflammatory responses by inhibiting the activation of the PI3K/Akt1/NFκB signaling pathway in a NFκB signaling-mediated autophagy way.PMID:34464886 | DOI:10.1016/j.intimp.2021.108096
