Micro‐RNA‐30a regulates ischemia‐induced cell death by targeting heat shock protein HSPA5 in primary cultured cortical neurons and mouse brain after stroke
Micro‐RNAs (miRs) have emerged as key gene regulators in many diseases, including stroke. We recently reported that miR‐30a protects N2A cells against ischemic injury, in part through enhancing beclin 1‐mediated autophagy. The present study explores further the involvement of miR‐30a in ischemia‐induced apoptosis and its possible mechanisms in primary cortical neurons and stroked mouse brain. Increases in intracellular calcium perturb blood–brain barrier via protein kinase C-alpha and apoptosis
This study reveals successive increases in intracellular calcium levels and protein kinase C-α activity are key mechanisms in OGD±R-mediated impairment of BBB. Furthermore inhibition of protein kinase C-α may be therapeutic in restoring BBB function by reducing the rate of cytoskeletal reorganisation, oxidative stress and apoptosis. Graphical abstract

Cytosolic phospholipase A 2 plays a crucial role in ROS/NO signaling during microglial activation through the lipoxygenase pathway
Conclusions: In summary, the results in this study demonstrated the role of cPLA 2 in microglial activation with metabolic links to oxidative and inflammatory responses, and this was in part regulated by the AA metabolic pathways, namely the LOXs. Further studies with targeted inhibition of cPLA 2 /LOX in microglia during neuroinflammatory conditions can be valuable to investigate the therapeutic potential in ameliorating neurological disease pathology.

Brain protection against ischemic stroke using choline as a new molecular bypass treatment.
CONCLUSION: Choline effectively attenuates brain ischemic injury in pMCAO rats, possibly by facilitating pial arteriogenesis and cerebral-cortical capillary angiogenesis via upregulating α7 nAChR levels and inducing the expression of HIF-1α and VEGF. siRNA mediated down-regulation of Sprouty2/4 diminishes ischemic brain injury
In this study, siRNAs directed against Sprouty2 and -4 were stereotactically injected along with the vasoconstrictive peptide endothelin-1 to create cortical infarcts in the adult rat brain. A single injection of Sprouty2/4 siRNAs (25μM each) significantly decreased Spry2 and Spry4 mRNA levels two days later and diminished the size of the injury area in the subchronic phase following vasoconstriction. Andrographolide stimulates p38 mitogen-activated protein kinase-nuclear factor erythroid-2-related factor 2-heme oxygenase 1 signaling in primary cerebral endothelial cells for definite protection against ischemic stroke in rats.
In conclusion, andrographolide increased Nrf2-HO-1 expression through p38 MAPK regulation, confirming that it provides protection against MCAO-induced brain injury. GPER expressed on microglia mediates the anti‐inflammatory effect of estradiol in ischemic stroke
ConclusionsOur studies have suggested that GPER expressed on microglia mediated the anti‐inflammatory effect of estradiol after ischemic stroke. Our studies could potentially help to develop more specific drugs to manage inflammation postischemic stroke. GPER was highly expressed in activated microglia after ischemia and estradiol. The specific GPER agonist G1 could inhibit the secretion of proinflammatory cytokines including IL‐1β and TNF‐α, which the anti‐inflammatory effect of G1 and E2 were both abolished by the specific GPER antagonist G15.

The Role of Staufen1 in Aberrant RNA Metabolism in SCA2 (P6.396)
Conclusions: Our results unravel a novel function for Staufen1 in aberrant RNA processing events and indicate its role in SCA2 pathogenesis. Our results further support a role for aberrant RNA metabolism in neurodegeneration thereby revealing its potential as a therapeutic target.
